Objective: To observe protective effect on DNA of retinal cone and rod cell of electric stimulation cerebellar fastigial nucleus on ischemic and reperfusion injury of rat retina at different ischemic time points.Methods: Rats were divideded randomly into group of ischemia reperfusion, group of electric stimulation treated and group of sham treated. Different ischemic time points are 0.5h, 1h, 2h, 3h, 4h, 5h, 6h. Reperfusion time is 6 hours. Electric stimulation is the first time of reperfusion for an hour. The group of sham treated is only an animal model of electric stimulation, but not be stimulated. Expression of injury in retina was measured by single-cell gel electrophoresis. Caspase-3 was measured by immunohistochemistry.Results: OliveTaliMoment of group of electric stimulation is significantly less than one of group of others except 0h and 0.5h. In every group, OliveTaliMoment is increased significantly at different ischemic time points, but from 3h to 4h is almost identical. OliveTaliMoment means DNA damage. Overall, OliveTaliMoment is increasing with ischemic time extended in every group, but OliveTaliMoment in 0.5h is small than one in 0h.Expression of Caspase-3 is positive in retinal cone and rod cell of all groups except 0h. Expression of Caspase-3 in group of ischemia reperfusion and group of sham is more significantly than in group of electric stimulation treated, but group of ischemia reperfusion is almost identical with the group of sham in every time point. But in 6h, group of electric stimulation treated express more significantly than the rest groups. Overall, optical density value of Caspase-3 is increasing from 0h to 3h, almost identical from 3h to 4h and decreasing 3h to 6h.Conclusions: (1) DNA of retinal cone and rod cell can be protected by electric stimulation cerebellar fastigial nucleus in limited time. (2)It is the valid period of treatment before 4h ischemic and best period before 2h.
