Objective: Researching miR-29C regulates Bak1in the process ofischemia-reperfusion injury rat model induced by electrical stimulation ofcerebellar fastigial nucleusMethods: Adult male SD rats (n=60), weight(280-300g), were divided into3groups randomly: pre-electrical stimulation group, simple model group, shamoperation group pre-electrical stimulation group was electrical stimulationed1hour, after24hours of the stimulation MCAO was performed andreperfusioned2hours later,and24hours later executed. infarction volumes weremeasured following TTC staining,animal behavior assessed the neurologicimpairment,and AOI was detected by TUNEL staining,chip analysis searchedthe the biggest expression differences miRNA in the groups,RT-PCR testified theexpession of this miRNA,and predicted it ’ s target gene through thebioinformatics software,at last we used of the dual-luciferase assay system toverify the relation between the miRNA and geneResults:In the pre-electrical stimulation group and simple model group the the volume of infarction were37.0±8.9%and62.0±10%(P<0.01), the AOI inexperimental groups were23.4±2.4%and58.9±4.0%(P<0.01),animal behaviorassessment were1.5±0.52and2.4±0.51(P<0.01)chip analysis found themiR-29C is the biggest expression differences miRNA in the2groups, RT-PCRshowed that miR-29C was down–regulated in pre-electrical stimulation groupcompared with simple model group,and the bioinformatics software predictedBak1is a target gene of miR-29C,in the dual-luciferase assay system thefluorescence value in Bak1-wt/rnomiR-29C groups was the lowest in the4groups which were Bak1-wt/NC group、 Bak1-wt/rno-miR-29C group、Bak1-Mut/NC group and Bak1-Mut/rno-miR-29C group(P<0.01),and thesewas no significant differences in the other3groups (P>0.05)Conclusion: Electrical stimulation of cerebellar fastigial nucleus in ratalleviated the cerebral ischemia-reperfusion injury;pre-electrical stimulationcerebellar fastigial nucleus down–regulates miR-29C;Bak1is a gene ofmiR-29C; whether miR-29C regulates Bak1in the mechanism of ischemiccerebral protection induced by electrical stimulation needs further vivovalidation.
