Objective:To investigate the expression of KAI1 protein,nm23 protein and micro-lymphatic vessel density(MLVD) in laryngeal squamous cell carcinoma(LSCC),combined with clinical data,such as clinical stage,histological grade and lymphatic metastasis in order to explore relationship between the expression of KAI1 protein,nm23 protein and lymphangiogenesis,lymphatic metastasis and Lymphangiogenesis and its role in occurrence,development and metastasis of laryngeal cancer.Methods:The expression KAI1 protein,nm23 protein were detected by immunohistochemistry(IHC) in 45 cases with laryngeal squamous cell carcinoma and 22 cases with normal mucosa tissues of larynx.Lymphangiogensis was evaluated by IHC with the specific antibody D2-40.All the data including clinical data were analyzed statistically.Results:1.the expression of KAI1 protein:(1) The expression of KAI1 protein was lower in tumor tissue than in normal tissue(P<0.05).(2) The expression of KAI1 protein was lower in LSCC with lymphatic metastasis than in LSCC without lymphatic metastasis(P<0.05). There was no significant association between the expression of KAI1 protein and T stage, grade of differentiation,age,gender(P>0.05).2.the expression of nm23 protein:(1) The expression of nm23 protein was lower in tumor tissue than in normal tissue(P<0.05).(2) The expression of nm23 protein was lower in LSCC with lymphatic metastasis than in LSCC without lymphatic metastasis(P<0.05).There was no significant association between the expression ofnm23 protein and T stage,grade of differentiation,age,gender(P>0.05).3.the count of MLVD:(1) MLVD was higher in tumor tissue than in normal tissue(P<0.01).(2) MLVD was higher in LSCC with lymphatic metastasis than in LSCC without lymphatic metastasis(P<0.01).4.In 45cases with LSCC,the MLVD was respectively lower in KAI1 and nm23 protein positive groups than those in the negative groups(P<0.05).Conclusion:1.The expression of KAI1 protein and nm23 protein was lower in tumor tissue than in normal tissue,as well as in LSCC with lymphatic metastasis than in LSCC without lymphatic metastasis,respectively(P<0.05),which may imply the lower expression of KAI1 protein and nm23 protein play a roles in occurrence,development and metastasis of LSCC.2.The lower expression of KAI1 protein and nm23 protein are related to lymphatic metastasis,which may be a useful indicator of potentiality of lymphatic metastasis in LSCC and provide evidence for the choice of clinical treatment programs and the assessment of prognosis.3.MLVD was higher in tumor tissue than in normal tissue and was related to lymphatic metastasis,which showed that lymphangiogenesis occurred in the development of LSCC.4.The lower expression of KAI1,nm23 protein in LSCC may play an role in the process of Lymphangiogensis.
