Expression And Clinical Significance Of High Molility Group Protein Box-1 (HMGB1) In Laryngeal Squamous Cell Carcinoma

Purpose:To investigate the expression situation of HMGB1 in laryngeal squamous cell carcinoma(LSCC) tissue and serum specimens and approach its significance in LSCC.Methods:Semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) and western blotting (WB) were respectively performed to measure messenger RNA and protein of HMGB 1 relative expression in 30 pairs cancerous,adjacent cancerous tissues(more than 5mm from cancerous tissues) paired samples. Enzyme-linked immunosorbent adsorption experiment(ELISA) was used to detect the expression situation of HMGB 1 protein in LSCC patients'serum.Results:Gray scale value are used to show the express of HMGB 1 mRNA and protein.From the results of RT-PCR we obtained the mean±standard deviation(x±SD) for HMGB 1 mRNA (HMGB1/GAPDH) in 30 LSCC tissues is 1.25±0.12 compared with 0.32±0.04 in 30 adjacent cancerous tissues. There was statistical significance between them (P<0.05). From the results of western blotting(WB) we obtained the mean±standard deviation (x±SD) for HMGB1/β-Actin in 30 pair LSCC tissues and adjacent cancerous tissues are 1.29±0.10; 0.34±0.03.There was significant difference (P<0.05). Statistical analyses of the two results above displayed the expression of both HMGB1 mRNA and HMGB1 had nothing to do with age, drinking and the location and pathology classification of tumor (P>0.05), but associated with smoking, T,N and clinical stages of LSCC (P<0.05). From the results of ELISA we obtained the mean±standard deviation (x±SD) for HMGB1 protein expression in 30 LSCC serums is 24.80±14.08(ng/ml) compared with 23.58±14.69(ng/ml) in 10 healthy human serums. There was no significant difference (P>0.05).Conclusions:The results in our studies suggested that HMGB1 mRNA and HMGB1 protein were overexpressed in LSCC tissue and associated with smoking, T,N and clinical stages of LSCC (P<0.05).We supposed they may be involved in the malignant transformation of HMGB 1.The strong correlation between the expression of HMGB1 mRNA and protein and pathological parameters of LSCC indicated HMGB1 may play a part in the invasion,metastasis,progression of LSCC. The expression of HMGB 1 protein in serum was no significant difference between LSCC patients and the healthy.
