The honeysuckle is clinical traditional Chinese medicine in common use.It has the effecting of removing heat and toxic substances,and dispeling wind-heat.As natural nutrient-rich health food for human beings,propolis is an important source of natural antioxidants due to its rich phenolic compounds.In this paper,antioxidant activities of honeysuckle extracts were determined in vitro systems,and chlorogenic acid and cafeic acid in honeysuckle were determinated using HPLC-ECD method. Furthermore,HPLC and HPLC-ESI-MS were used for evaluation of scavenging activity of propolis on 1,1-diphenyl-2-picrylhydrazyl(DPPH).This paper consists of five chapters,including four aspects as following:1.The antioxidant properties of honeysuckle extracts were evaluated using different methods including reducing power,iron(â…¡)chelation,DPPH radical and hydroxyl radical.The results showed that a positive correlation was observed between chlorogenic acid and the reducing power assay,hydroxyl radical assay.Ethanolic and methanolic extracts from honeysuckle showed higher antioxidant activities than water extracts.But water extracts from honeysuckle showed higher antioxidant activities than Ethanolic and methanolie extracts in DPPH radical assay and iron(â…¡)chelation assay,which indicated that honeysuckle contained other antioxidation components except chlorogenic acid.2.To establish a high performance liquid chromatography with electrochemical detection(HPLC-ECD)method for determination of chlorogenic acid and cafeic acid in honeysuckle.The results showed that two kinds of phenolic acids were separated satisfactorily.The sensitivity of chlorogenic acid and caffeic acid,obtained with ECD analyses,were 4.1,11.6 times,respectively,higher than those obtained by DAD. This study might provide a scientific basis for the quality control of honeysuckle.3.A rapid and accurate HPLC-UV method for determination of scavenging activity of propolis on 1,1-diphenyl-2-picrylhydrazyl(DPPH)has been developed. The results showed that DPPH eluted as a sharp peak at 3 min or so.Compared with the colorimetric method,the HPLC method was accurate,specific and free from color pigments interferences from propolis.4.The main antioxidant components in propolis were studied by HPLC-ESI-MS method.The results showed that the dissociation laws of some antioxidant componts in propolis.This study not only identified the antioxidant components in propolis but also provided the structure information of these antioxidant components...
