Purpose:To investigate the expression and correlativity of Smad4 and P21 WAFI/CIPI in benign epithelial ovarian tumours and borderline epithelial ovarian tumours and epithelial ovarian cancer separately,to provide the experimental basis for researching the development of epithelial ovarian cancer。Methods:To detect the expression of Smad4 and P21 WAFI/CIPI in 24 tissues of benign epithelial ovarian tumours.22 tissues of borderline epithelial ovarian tumours,43 tissues of epithelial ovarian cancer by Eli vision immunohisto-chemical method.Results:1. The positive expression of Smad4 was 70.8%in benign epithelial ovarian tumours,81.8%in borderline epithelial ovarian tumours and 81.3%in epithelial ovarian cancer. Pairwise comparison between the three groups had no significantly correlated(P>0.05). The expression rate of Smad4 protein in epithelial ovarian cancer had no correlated with different clinical stage, different tumor histologic types,different pathological types and whether lymph node metastasis(P>0.05).2. The positive expression of P21 WAFI/CiPI Was62.5%in benign epithelial ovarian tumours,46.6%in borderline epithelial ovarian tumours and 34.9%in epithelial ovarian cancer.The positive expression rate of P21WAFI/CIPI protein was not significantly correlated between the benign epithelial ovarian tumour group and the borderline epithelial ovarian tumour group(P>0.05).The positive expression rate of P21 WAF1/CIPI protein was significantly correlated between the borderlin epithelial ovarian tumour group and the epithelial ovarian cancer group(P<0.05).And the expression rate of P21 WAFI/CIPI protein was significantly correlated between the benign epithelial ovarian tumour group and the epithelial ovarian cancer group(P<0.05).And it had significantly difference with tumor different clinical stages and pathological types(P<0.05). The later clinical stage, the lower P21 WAFI/CIP1 positive rate. The positive expression rate of P21 WAFI/CIPIis higher in the well-differentiated epithelial ovarian cancer than the poorly differentiated epithelial ovarian cancer. But no correlated with different tumor histologic types and whether lymph node metastasis(P>0.05).3. In epithelial ovarian cancer, Smad4 protein and P21WAFI/CIPI protein expression was no significant correlation (Rs=0.099,P>0.05).Conclusion:1. In the process of ovarian tumor from benign to malignant transformation, the positive expression of Smad4 protein was no significant difference. In ovarian cancer tissue, the position experssion of Smad4 protein was not correlated with the clinical and pathological features. These data suggest that the absence or experssion of Smad4 protein is not the key genes leading to the canceration of ovarian epithelium.2. In the process of ovarian tumor from benign to malignant transformation, the positive expression of P21WAFI/CIPI protein was decreased. In ovarian cancer tissue, the worse the pathological differentiation and the later clinical stage, the p21WAF1/CIPI protein position expression rate is lower. These data suggest that the absence of P21WSFI/CIPI is closely related with the malignant progression of ovarian tumour.3. In ovarian epithelial carcer,the expression or absence of P21WAFI/CIPI protein is out the control of Smad4 protein.
