Objective to assess the effect of calcium dobesilate on the protection against diabetic nephropathy and on the expression of intercellular adhesion molecular-1(ICAM-1) and vascular cell adhesion molecular-1(VCAM-1) in diabetic rat kidney.Methods The rats model of diabetic mellitus (DM)was produced by injection of streptozocin (STZ) after nephrectomy. Eighteen rats were assigned to normal control(C)group,diabetic rat(DM)group,calcium dobesilate treatment(R)group. Immunohistochemical analysis,Western blot were used to examine the pathological change of kidney and expression of ICAM-1 and VCAM-1 after 12 weeks in the kidney of rats.Result 1)After 12-week treatment,compared with diabetic rat(DM)group, treatment group exhibited that,Scr and urine protein decreased( P<0.05 ) ,Ccr increased (P<0.05).2)histopathology of kidney indicated that ,the glomerulosclerosis index of treatment group decreased (P<0.05). 3)In comparison with normal group , the expression ICAM-1 and VCAM-1 in diabetic rat and treatment group was markedly increased (P<0.05). In comparison with diabetic rat group, calcium dobesilate could significantly decrease the expression of ICAM-1 and VCAM-1(P<0.05).Conclusion Calcium dobesilate may play a protective role of the kidney by down-regulating ICAM-1 and VCAM-1 ,thus preventing the progression of diabetic nephropathy.
