【Objects】To optimize the extraction method of polysaccharide in Coptis chinensis(CPS). Study on antioxidant activity, hypoglycemic activity and antibacterial activity of CPS in vitro.【Methods】Water-extract & alcohol-precipitate method was adopted to extract CPS, uniform design method was used to optimize the extract method of CPS; inhibition ratio to·OH, O2-·and DPPH was used to appraise the antioxidant activity; inhibition ratio toα-amylase was used to appraise the hypoglycemic activity; agar diffusion method was adopted to appraise the antibacterial activity.【Results】①Gray CPS was procured and the content in Coptis chinensis is 0.32~0.81%. Optimized extract condition is at 100℃, 1:10(g/ml) of the solid-solvent ratio and 120 minutes.②Contrast the differences of CPS extract on different pH levels, the antioxidant activity experiment indicates that CPS extracted with water and use papain and Sevag method combined to deproteinization has better effect than extracted on other methods, it's EC50to·OH is 83.91μg/ml obviously surpass Manicol, and to DPPH is 249.86μg/ml obviously surpass VE, but it has quiet effect of inhibition O2-·.③The hypoglycemic activity experiment shows that CPS extracted with water and use papain and Sevag method combined to deproteinization also has an excellent activity of reduce blood glucose, the EC50 is 182.36μg/ml obviously surpass Acarbose.④CPS can not form a cingula on a beef extract & peptone agar culture with Bacillus coli or Staphylococcus aureus accreted, however, liquid extract of Coptis chinensis and berberine hydrochloride are effective.【Conclusions】①The content of CPS in Coptis chinensis is 0.32~0.81%. Optimized extraction condition shows that the temperature is the main factor to increase the yield of CPS.②Antioxidant activity experiments in vitro shows CPS can be antioxidant and a good free-radical scavenger.③Hypoglycemic activity experiments disclose CPS has an excellent activity of reduce blood glucose and obviously surpass Acarbose. CPS can be hoped to be medicine.④No antibacterial activity was fond through agar diffusion method.
