Objective:Centromere protein F(CENP-F) is an important mitotic apparatus protein.It is reported to be related to malignant proliferation of most of human tumors abroad.Vascular endothelial growth factor(VEGF) is the key factor of tumor vessel poietic.It is reported to be related to tumor vessea poietic abroad and home.We have previously reported that CENP-F is over expressed in human brain gliomas,and some of which are expressed in vascular endothelial cell of human brain gliomas.The aim of this article is to detect the expression of CENP-F in human brain gliomas,and to investigate the relationship among centromere protein F(CENP-F),Vascular endothelial growth factor(VEGF) the vascular proliferation and in glioma.Investigate the relevance of VEGF,CD34,CENP-F and several clinical factors with tumor malignant degree and prognosis.Methods:(1) Sixty formalin-fixed,paraffin-embedded gliomas were obtained from the Pathology Department of the First Affiliated Hospital of Anhui Medical University during the year of 2003 and 2005.All samples were examined by two senior pathologists and were confirmed as gliomas.At the same time,the clinical pathologic data of all patients were collected from the Patient's Condition Department of the First Affiliated Hospital of Anhui Medical University.Ten normal brain tissues(non-tumor brain tissues) were obtained as normal control from surgical resections of trauma patients,for whom a partial resection of normal brain tissue was required as decompression treatment for their severe head injuries to reduce increased intracranial pressure.The SP immunohistochemical staining method was used to detect the expression of VEGF and CENP-F in 60 cases of human gliomas of various pathological types and grades and 10 cases of non-tumor brain tissue samples.CD34 was used to examine the MVD of them,and analyze the relationship among MVD, VEGF and CENP-F.Investigate the relevance of VEGF,CD34,CENP-F and several clinical factors with tumor malignant degree and prognosis.(2) Follow-up was performed on 60 cases of gliomas by out-patient clinic,telephone and letters.(3) Analytical method:staistics analyses by SPSS11.0 software,quantitate data means use t test,classification data means useχ~2 test,categorization variance association analysis.the relevance of various kinds and prognosis use Logstic regression factor analysis.Results:(1) The expression of VEGF,CD34 and CENP-F were correlated with glioma malignant degree.CENP-F and VEGF were correlated with vascular proliferation in glioma.(2) The results of multivariate logistic analysis showed that the malignant degree,the preoperative karnofsd prognosis score(KPS ),the VEGF and the CENP-F expression were significantly correlated with 3 annual survival rate.The malignant degree,the preoperative KPS and CENP-F expression were prognostic markers of glioma. Conclusions CENP-F and VEGF were correlated with vascular proliferation in glioma. The expression of VEGF,CD34 and CENP-F were correlated with glioma malignant degree.The preoperative KPS score and the CENP-F expression were significantly correlated with prognosis.The malignant degree,the preoperative KPS score and CENP-F expression were prognostic markers of glioma.MeSH Glioma / brain neoplasms / Vascular endothelial growth facto / prognosis.
