Objective:To investigate the expression of transcription factor Sp1,KLF6,c-fos expression in hepatocellular carcinoma (HCC) and liver cirrhosis tissue, and explore their functional mechanisms in the oncogenesis and development of HCC and liver cirrhosis. TGFβ1 and Ketanserin were used to stimulate HepG2 cell,the expression of HepG2 cell Sp1mRNA was detected and in order to preliminarily explore the functional mechanisms of Sp1 in this process.Methods:Using immuno-histochemistry, we studied the Sp1,KLF6,c-fos protein expression in normal liver,liver cirrhosis,para-carcinoma and HCC tissues specimens that obtained from patients who underwent liver diseaseas. The expression of HepG2 cell Sp1mRNA was detected with RT-PCR method after the interference of TGFβ1 and Ketanserin.The apoptosis rate of HepG2 cell was detected with FCM after the interference of TGFβ1 and Ketanserin.Results:The expression of Sp1,KLF6,c-fos protein obviously increased in HCC tissue and para-carcinoma tissue,compared with normal liver tissue (P<0.05);c-fos protein expression in liver cirrhosis tissue is high, compared with normal liver tissue(P<0.05);Sp1 and KLF6 protein expression in liver cirrhosis tissue is weak, compared with normal liver tissue(P>0.05).With the increasing concetration of TGFβ1 and Ketanserin,the expression of HepG2 cell Sp1mRNA gradually decreased.Conclusion:1 The expression of Sp1,KLF6,c-fos protein obviously increased in the HCC ,implied that they were connected with occurrence of HCC.2 The expression of Sp1,KLF6 protein in liver cirrhosis is weak, we can not find that they were connected with occurrence of liver cirrhosis.3 The expression of c-fos protein in liver cirrhosis is strong, implied that it was connected with occurrence of liver cirrhosis .4 TGFβ1 and Ketanserin could induced the apoptosis of HepG2 cell,the decreased expression of Sp1 mRNA may be one mechanism of it.
