Objective:To investigate the mechanism of Iodination-125 refraining the growth of tumors cell by hindering the expression of Tyro-sine Kinase Protooncogene A(TrKA)and Basic Firoblast Growth factor(bFGF)in breast cancer.Methods:The animal models of the Breast cancer cell(MCF-7)were established by subcutaneously injecting the tumor cells into nude mice,then,models were randomly grouped and the Iodine-125 granule or no-load seeds were implanted into the tumor mass according:to the division of groups.The mice were divided into eight groups and there were six nude mice in each group;the control groups contained D7,D14,D21,D28 groups;the experiment groups contained S7,S14,S21,S28 groups.No-load seeds,non-iodine-125 radioelement(0mCi)were implanted into the control groups;Iodine-125 seeds(0.4mCi)were implanted into the experiment groups,when the length of tumor was about 8-10mm.The size of tumors was measured every two days and their volumes then were calculated.Anatomy analysis were done separately after 1,2,3and 4 weeks of the implantation of the Iodine-125 seeds;the samples were frozen and fixed to preserve according to different groups;the mRNA and proteinum expressions of TrKA and bFGF were measured by the ways of RT-PCR and immunohistochemistry.Results:Our experiment had shown that after Iodine-125 seeds were transplanted into the tumors of the experiment groups,tumors grew more slowly,the tumor inhibitory rate was 55.27%and it obviously rose compared with the other groups.The mRNA and proteinum express level of TrKA and bFGF in tumors were significantly decreased in experiment groups. There was significant difference between the two groups(P<0.01).TrKA and bFGF expressions of tumor tissue were positive correlation(P<0.05)in level of protein and gene.Conclusions:It is one of important molecular biology mechanisms to restrain breast cancer with iodine-125 seeds to decreass the expression of TrKA and bFGF in tumor cells.
