Part 1:The effect of salvianolate on the focal cerebral ischemia-reperfusion injury in ratsObjective: to detect the protective effect of Salvianolte on the focal cerebral ischemia-reperfusion injury in rats and to reveal the mechanism.Methods: 48 SD rats were divided into four group randomly, normal control group, ischemia control group, salvianolate(10 mg/kg) group and salvianolate(30 mg/kg) group, each group contein 6 rats. We used the Longa method to establish the MCAO model. The salvianolte at different dose were injected introperitoneally at the different time point according to the test design. The brain infarct volume by TTC stain and behavior scores by Long's methods were also determined in order to evaluated the protective effect. Also we use assay kit to determine the activities of SOD and GSH-Px, content of MDA and NO in each group.Results: (1): The activities of SOD and GSH-Px of brain tissue in the ischemia control group, salvianolate(10 mg/kg) group and salvianolate(30 mg/kg) group are much lower than those in normal control group. But the salvianotes can inhibit this decrease.(2): The content of MDA of brain tissue in the ischemia control group, salvianolate(10 mg/kg) group and salvianolate(30 mg/kg) group are much higher than those in normal control group. The salvianolate can inhibit this increase.(3): The content of NO of brain tissue in the ischemia control group, salvianolate(10 mg/kg) group and salvianolate(30 mg/kg) group are much higher than those in normal control group. The salvianolate can inhibit this increase.Conclusion: The salvianolate plays neuropective effect maybe through modulating the imbalance of oxidant and antioxidant inducing by the cerebral ischemia-reperfusion. Part 2:The effect of salvianolate on the expression of BDNF and GDNF protein of the brain tissue following focal cerebral ischemia-reperfusion in ratsObjective: to observe the effect of salvianolate on the expression of BDNF and GDNF protein of the brain tissue following focal cerebral ischemia-reperfusion in rats, and to reveal the mechanism of neuroprotective effect of salvianolate.Methods: We use ELISA method to detect the the expression of BDNF and GDNF protein of the brain tissue in each group.Results: The expression of BDNF and GDNF protein of the brain tissue in ischemia control group are much higher than those in normal control group. The expression of BDNF and GDNF protein in salvianolate(10 mg/kg) group and salvianolate(30 mg/kg) group even much higher than those in ischemia control group.Conclusion: The salvianolate plays neuropective effect maybe through increase the BNDF and GDNF protein in the brain tissue following the cerebral ischemia reperfuison injury.
