Part one: Effects of intrathecal Ro25-6981 in rat model of middle cerebral artery occlusion on neurological deficit scoreObjective To explore the effection of intrathecal injection Ro25-6981 in rat model of middle cerebral artery occlusion on neurological deficit scoreMethods 48 male Sprague-Dawley rats weighed 280-320g were randomly divided into four groups: (1)sham group, (2)ischemic-reperfusion group ( I/R) ,(3)Ro25-6981 treatment group (I/R+Ro),(4) NS treatment group (I/R+NS).Each group had 12 animals. The middle cerebral artery in rats was occluded for 2h by an intraluminal filament and then reperfused to cause transient focal cerebral ischemia. In the I/R+Ro group, the Ro25-6981 was injected intrathecally 22h after reperfusion. In the I/R+NS group, the Normal saline was injected intrathecally 22h after reperfusion.Grading the neurological deficit score 24h after reperfusion.Results The neurological score in I/R group were higher than sham group,the results were statistically significant difference from sham group( P < 0.01 ). The neurological score in I/R+Ro group was lower than I/R+NS group,the results were statistically significant difference from I/R+NS group ( P < 0.01 ).Conclusion The results suggest that inject Ro25-6981 intrathecally 22h after reperfusion in rat model of middle cerebral artery occlusion can reduce neurological score. The infarct volum in I/R+Ro group was lower than I/R+NS group,the results were statistically significant difference from I/R+NS group ( P < 0.05 ).Conclusion The results suggest that inject Ro25-6981 intrathecally 22h after reperfusion in rat model of middle cerebral artery occlusion can reduce infarct volum. |
