Objective: To investigate the expression of CD44v6, MMP-9 and nm23-H1 protein in cholangiocarcinoma and normal bile duct tissues, in order to find out the role in the invasion and metastasis of cholangiocarcinoma and try to clarify the mechanism .Methods: In this study we have investigated the expression of CD44v6, MMP-9 and nm23-H1 protein in 55 cases of cholangiocarcinoma and 12 cases normal bile duct tissues by immunohistochemistry(PV-9000). The different expression levels in two kinds of specimens were dealed with statistical analysis, so were the correlation between the expression and the clinic pathologic character, as well as the correlation among the three.Results: (1) The percentage of the CD44v6-positive tissue was significantly higher in the cholangiocarcinoma (70.9%)than in the normal bile duct (16.7%),(P<0.01); The percentage of the MMP-9-positive tissue was significantly higher in the cholangiocarcinoma (83.6%)than in the normal bile duct (0%),(P<0.01); The percentage of the nm23-H1-positive tissue was significantly lower in the cholangiocarcinoma (54.6%)than in the normal bile duct (83.3%),(P<0.05). (2)The expression of CD44v6 protein in cholangiocarcinoma is significantly related with tumor invasion and metastasis and clinical stage of tumor according to TMN and not related with patient's age ,sex,tumor stage; The expression of MMP-9 protein in cholangiocarcinoma is significantly related with tumor stage, tumor invasion and metastasis and clinical stage of tumor according to TMN and not related with patient's age ,sex; The expression of nm23-H1 protein in cholangiocarcinoma is significantly related with tumor stage,tumor invasion and metastasis and clinical stage of tumor according to TMN and not related with patient's age, sex. (3) there was a positive linear correlation between the expression of CD44v6 and MMP-9 in cholangiocarcinoma (C=0.298, P<0.05); there was a negative linear correlation between the expression of CD44v6 and nm23-H1 in cholangiocarcinoma(C=0.325, P<0.05); there was a negative linear correlation between the expression of MMP-9 and nm23-H1 in cholangiocarcinoma (C=0.318, P<0.05).Conclusion: 1 The expression levels of CD44v6, MMP-9 and nm23-H1 proteins were significantly higher in cholangiocarcinoma than in normal bile duct. They have intensive correlation with the growth of cholangiocarcinoma.2 The expression of CD44v6, MMP-9 and nm23-H1 proteins in cholangiocarcinoma were significantly related with tumor invasion and metastasis. These results showed that the expression of them were significantly related with cholangiocarcinoma invasion and metastasis. 3 The expression levels of CD44v6, MMP-9 and nm23-H1 proteins may be in coordination with the development of cholangiocarcinoma.
