Expressions And Significance Of TNF-α, CGRP In The Portal Hypertensive Rat's Stomach, Small Intestine, Colon Tissue | Posted on:2009-01-08 | Degree:Master | Type:Thesis | Country:China | Candidate:K Li | Full Text:PDF | GTID:2144360245469009 | Subject:Surgery | Abstract/Summary: | PDF Full Text Request | Objective: To observe portal hypertensive rat model of TNF-α, CGRP in portal hypertensive rats stomach, small intestine and colon tissues, and to explore its significance in portal hypertensive gastropathy and portal hypertension enteropathy. Methods: Male Sprague-Dawley rats(SD rats) were randomly divided into the experimental group and control group , the experimental group performed the two-stage ligation of portal vein plus ligation of the left adrenal vein, Two weeks after complete portal ligation, portal vein pressure was measured. The levels of tumor necrosis factor -α(TNF-α) and calcitonin gene-related peptide (CGRP) in stomach, small intestine and colon tissue were detected by immunohistochemistry. Results: 1. Two weeks after complete portal ligation, portal vein pressure in the experimental rats (21.85±3.57cmH2O) was significantly higher than in controls(12.03±1.85cmH2O), (P <0.05).2. In stomach tissue , TNF-αand CGRP in experimental rats showed strong positive expression, and the control group were negative and weakly positive expression.3. In small intestinal tissue , TNF-αand CGRP expression intensity in experimental rats was significantly higher than in controls.4. In colon tissue , TNF-αand CGRP expression intensity in experimental rats was significantly higher than in controls. Conclusions: 1. A rat model of portal hypertension can be established with two-stage ligation of portal vein plus ligation of the left adrenal vein.2. TNF-αand CGRP involved in the occurrence and development of portal hypertensive gastropathy (PHG) .3. TNF-αand CGRP involved in the occurrence and development of portal hypertension enteropathy(PHE).
