Objective:To study the expression of TK1 and Ki67 in breast tumor.To investigate the relationship between TK1 and Ki-67,and their correlation with ER,PR,C-erbB-2,P53,the clinical factors and pathological factors in breast cancer.To explor the effection of the expression of TKl,Ki67 and their unit in the metastasis and recurrence of breast cancer.Materials and method:Selected 65 cases which followed up from xiang ya Hospital Breast Department from March 1997 to July 2007 randomly. Put them in two groups including 37 cases with recurrence or metastasis in 5 years(group A),28 cases without recurrence or metastasis in 5 years(group B).Took other 8 cases with breast fibroid tumor as group C and 10 cases of normal breast tissue as group D.The expression of TK1 and Ki67 was detected in the four groupd by immunohistochemical staining technique.All data was analyzed by SPSS 13.0 for windows.Results:(1)The positive staining of TK1was mainly allocated in cytoplasma,partly in nucleus.The positive expression rate of TK1 in 65 patients with breast cancer was 81.5%(53/65).Only one express positive in 8 fibroid tumors and none in 10 normal breast tissues.There was significant difference between breast cancer,breast fibroid tumor and normal breast tissue(P<0.001).(2)The positive staining of Ki67 was allocated in nuleus.No cutoplasm and cytolemma was stained. The positive expression rate of Ki67 in 65 patients with breast cancer was 61.5%(40/65).No positive expression in in 8 fibroid tumors and 10 normal breast tissues. There was significant difference between breast cancer,breast fibroid tumor and normal breast tissue(P<0.001).There was no significant difference betweenTK1 and Ki67 in all 65 breast cancer cases(P=0.089).(3)The positive expression of TK1 was significantly and positively correlated with that of Ki67(r=0.250,P=0.046);and not correlated with that of ER,PR,C-ercB-2,P53and the clinical and the pathological factors such as age,neoadjuvant chemotherapy,etc(P>0.05).(4)The positive expression rate of Ki67 was significantly different from histological grade I to III(p=0.037);and and not correlated with that of ER,PR,C-ercB-2,P53and other clinical and the pathological factors(p>0.05).(5)The expression of TKl,Ki67and their unit between group A and B showed statistical difference(P=0.046,0.003, 0.023).The results of survival analysis showed that positive expression of both TK1 and Ki67represent worse prognosis than that not.Conclusions:The expression of TK1 was significantly higher in breast cancer than that of in fibroid tumor and normal breast tissue.TK1 was conduced to the judgement of breast cancer,but was not able to be a independent prognostic index.The expression of TK1 was significantly higher in breast cancer than that of in fibroid tumor and normal breast tissue. The positive expression rate of Ki67 was significantly different from histological grade I to III.Positive expression of both TK1 and Ki67 foreshowed a high reccurence or metabasis of breast carcinoma.
