Expression Significance Of BRCA1and EGFR In Endometrial Carcinoma

OBJECTIVETo investigate the expressions of BRCA1,EGFR in normalendometrium,endometrial atypical hyperplasia and endometrial cancer(EC).Toexplore their relationship with the occurrence and development of endometrialcancer.METHODSImmunohistochemical methods were used to detect the expression ofBRCA1and EGFR in normal endometrium,endometrial atypical hyperplasiaand endometrial cancer.The correlations of expression levels and endometrialcarcinoma clinicopathological characteristic were analyized with theage,menopause,stage,histopathology grade,lymph node metastasis,depth ofmyometrial invasion and follow-up data.RESULTS(1) We found that the positive rates of BRCA1expressions in normalendometrium,endometrial atypical hyperplasia and endometrial cancer sampleswere90％(36/40),66.7％(20/30),31.7％(19/60). the difference wassignificant(p<0.05).The positive rates of EGFR expressions in normal endometrium,endometrial atypical hyperplasia and endometrial cancer sampleswere30％(12/40),36.7％(11/30),73.3％(44/60),the difference wassignificant(p<0.05).(2)BRCA1was associated with the age,menopause,stage, histopathologygrade,lymph node metastasis and depth of myometrial invasion (p<0.05). Theexpression raw was lower in the higher histopathology grade (p<0.05).(3) EGFR was associated with menopause,stage,the depth of myometrialinvasion and lymph node metastasis(p<0.05),but was not related to the age orhistopathology grade (p>0.05).(4) There was a statistically negative relationship between the expressionsof BRCA1and EGFR(r=-0.319,P=0.013＜0.05)．(5) The Kpan-Meier analysis showed that the BRCA1,EGFR andhistopathology grade were related with the prognosis of endometrialcancer(p<0.05).(6) The multivariate analysis Cox proportion hazards regression modelshowed that the EGFR ang histopathology grade was related with the prognosissignificantly (p<0.05).CONCLUSIONS1.BRCA1is low expression in endometrial carcinoma.The expression wasassociated with stage, histopathology grade,the depth of myometrial invasion,lymph node metastasis,and it may play an important role in the prediction formalignancy and prognosis of endometrial cancer.2.EGFR is high expressed in endometrial carcinoma.The expression wasassociated with stage,the depth of myometrial invasion,lymph nodemetastasis,and it is expected to be a useful marker for the prognosis ofendometrial cancer. 3.There is a negative correlation between the expression of BRCA1andEGFR, the BRCA1positive expression was lower in the higher EGFR positiveexpression.They are related to the progression of malignant endometrial cancerand poor prognosis;BRCA1and EGFR co-expression may predict poorprognosis and instruct postoperative therapy.
