The Expression And Significance Of Survivin And BcL-2 Protein In Breast Cancer

Objective:Breast cancer is the highest incidence rate malignant tumors in feminine disease at present,its occurrence has the trend of upgrade and young ,the occurrence of breast cancer and other tumors is a complicated physiological progress that multigene participated and multistep,during the progress apotosis inhibition plays a very important roles in the occurrence and development .Aim of this study is to observe the expression of Survivin and BcL-2 protein which is in the family of inhibitor of apoptosis in breast cancer and normal tissue by immunohistochemical method(PV method). The relationship between the expression of Survivin and BcL-2 is analyzed with clinical,pathological characteristics,such as pathological stages,tumour size ,lymph node metastasis condition and etc in breast cancer ,the correlation trend is investigated between the expression of Survivin and BcL-2,these provide evidence for breast cancer of early diagnosis and prognostic evaluation.Methods : 1 48 paraffin blocks of surgery female mammary gland were randomly selected from the He Bei province general hospital from august of 2006 to august of 2007,including 24 mammary infiltrating ductal carcinoma,14 mammary infiltrating lobular carcinoma and 10 mammary normal tissues.Clinical and pathological data of all patients were collected. The expression of Survivin and BcL-2 were detected by immunohistochemical method,the results were evaluated by the method of semiquantitative scores as Fromowitz [1].The difference between expression of Survivin and BcL-2 was compared in infiltrating mammary cancer and normal tissues,the relationship of Survivin and BcL-2 was analyzed with clinical,pathological characteristics of tumor,the trend of Survivin and BcL-2 was also analyzed.2 Statistical method:Use the SPSS16.0 software system to analyze the results,applicate the rank sum test,χ2 test and Spearman grade-related test,analyze the relationship between the expression of Survivin and BcL-2 clinical,pathological characteristics.Result:1 The positive rate of the expression of Survivin in breast cancer 86.84%(33/38) is higher than that in normal tissue10% (1/10),the difference has statistical significance(P=0.00).The positive rate of the expression of Bcl-2 in breast cancer65.79% (25/38) is higher than that in normal tissue 20.00%(2/10),the difference has statistical significance(P=0.00).There is no statistical significance on positive rate of the expression of Survivin and Bcl-2 between infiltrating ductal breast cancer and infiltrating lobular breast cancer (P>0.05). 2 The positive rate of the expression of Survivin in the no lymph node matastasis group 73.33%(11/15)is lower than that in the lymph node matastasis group 95.65%(22/23)in breast cancer, the expression of Survivin has positive correlation with the lymph node matastasis ( P<0.05);The positive rate of the expression of Bcl-2 in the no lymph node matastasis group 60.00%(9/15)is lower than that in the lymph node matastasis group 69.57%(16/23)in breast cancer,but the expression of Bcl-2 has no correlation with the lymph node matastasis ( P>0.05).3 the positive rate of the expression of Survivin in the group of diameter≤2cm,> 2cm≤5cm,> 5cm are separately 91.67%(11/12),85.00%(17/20),83.33%(5/6), but there is no correlation between them (P>0.05);The positive rate of the expression of Bcl-2 in the group of diameter≤2cm,>2cm≤5cm,>5cm are separately 58.33%(7/12),70.00%(14/20),66.67%(4/6),but there is no correlation between them (P>0.05).4 the positive rate of the expression of Survivin in the pathology stage ofⅠ-Ⅱgroup84.62%(22/26)is lower than that in the pathology stageⅢgroup91.67%(11/12),but there is no correlation between them (P>0.05);The positive rate of the expression of Bcl-2 in the pathological stage ofⅠ-Ⅱgroup 69.23%(18/26) is higher than that in the pathological stagesⅢ-Ⅳgroup 58.33%(7/12) ,but there is no correlation between them (P>0.05). 5 There is no statistical significance in the expression of Survivin and Bcl-2 in the menopause group and the non-meno- pause group (P>0.05).6 The positive rate of expression of Survivin in the group ofⅠgrade,Ⅱgrade,Ⅲgrade are separately 93.33%(14/15),73.33%(11/15),100.00%(8/8),there is no correlation between them(P=0.16>0.05);The positive rate of expression of Bcl-2 in the group ofⅠgrade,Ⅱgrade,Ⅲgrade are separately 80.00%(12/15),73.33%(11/15),25.00%(2/8),there is inverse correlation between them(P=0.01<0.05).7 The positive rate of the expression between Survivin and Bcl-2 have no statistical significance (P>0.05).Conclusions:1 The expression of Survivin and Bcl-2 is differenced in breast cancer.It is a a high expression of Survivin and Bcl-2 in breast cancer,and a low expression of in normal tissue. 2There is no correlation between Survivin and tumor diameter,pathological stage,menstruation condition,histology grade,but there is positive correlation between the expression of Survivin and lymph node matastasis condition.3 There is no correlation between Bcl-2 and tumor diameter,lymph node matastasis,pathological stage,menstruation condition, but there is inverse correlation between the expression of Bcl-2 and histology grade.4 There is no correlation between the expression of Survivin and BcL-2 in breast cancer.
