Objective To study the proliferative effects of NS398,a selective COX-2 inhibitor,on renal carcinoma cell line 786-O Which were treated with different concentrations of NS398 in Vitro. To detect the protein and mRNA expressions of COX-2 and showed the expressions of COX-2 were significantly inhibited by NS398. To study the apoptosis effects on renal carcinoma cell line 786-O Which were treated with different concentrations of NS398.Methods 786-O cells were treated with different concentrations of NS398 for 24h,48h,72h and 96h.the proliferation of 786-O cells treated with different concentrations was studied by MTT assay. Immuno cytochemistry was used to detect the expressions of COX-2 of 786-O cells treated with different concentrations. The expression of COX-2 mRNA of 786-O cells treated with different concentrations was analyzed by reverse transcription-polymerase chain raction(RT-PCR).Flow cytometric analysis was used to detect the apoptosis status of 786-O cells treated with different concentrations. Result NS398 result in significant antiproliferative effects on 786-O cells in dose and timedependent. RT-PCR showed the expressions of COX-2 were 0.482±0.12 and 0.266±0.39 in the 786-O cells treated with NS398 in different concentrations of 30μmol/L and 180μmol/L,which was significantly higher than that in the control group 0.776±0.035. Immunocytochemistry showed the expressions of COX-2 were 308±52 and 176±39 in the 786-O cells treated with NS398 in different concentrations of 30μmol/L and 180μmol/L,which was significantly higher than that in the control group 431±23. Apoptosis rate was 31.5±2.1% and 14.3±1.4% in the 786-O cells treated with NS398 in different concentrations of 180μmol/L and 30μmol/L,which was significantly higher than that in the control group 2.1±0.4%.Conclusion NS398 could effectively suppress the growth of 786-O cells,the mechanism probably relates to its inhabited expression of COX-2 and induce apoptosis.
