| ObjectiveTo investigate the therapeutic effect of Xiaoyu hematostatic tabella(XYHT) on blood clotting disorder,and approach the possible mechanism of it.MethodPart one Effect of XYHT on the platelet count and functoin and the blood clotting system of wista rats and mice(1).Normal mice were divided into 5 groups(high-dose group,midst-dose group,low-dose group,negative control group,possitive control group) randomly.The negative control group was treated with equal volume normal sodium(NS),possitive control group was treated with Yunnan white powder.Flowing intervention,bleeding time(BT),bleeding volum and coagulation time were analyzed.(2).Normal vista rats were divided into 5 groups(high-dose group,midst-dose group, low-dose group,negative control group,possitive control group) randomly.The negative control group was treated with equal volume normal sodium(NS),possitive control group was treated with Yunnan white powder.Flowing intervention,platelet count and functoin and thrombin time (TT),partial thromboplastin time(APTT),prothrombin time(PT) and Fibrinogen(Fg) were analyzed.Part two Effect of XYHT on the platelet count,functoin and the blood clotting system of blood coagulation dysfunction mice and rats model led by Warfarin(1).Blood coagulation dysfunction mice model were erected by Warfarin,and divided into 5 groups(high-dose group,midst-dose group,low-dose group,model group,possitive control group) randomly.Possitive control group was treated with Yunnan white powder,in addition,a negative normal mice control group which was treated with equal volume normal sodium(NS) was set up.Flowing intervention,BT,bleeding volum and coagulation time were analyzed.(2).Blood coagulation dysfunction vista rats model were erected by Warfarin,and divided into 5 groups(high-dose group,midst-dose group,low-dose group,model group,possitive control group) randomly.Possitive control group was treated with Yunnan white powder,in addition,a negative normal rat control group which was treated with equal volume normal sodium(NS) was set up.Flowing intervention,platelet count,functoin,TT,APTT,PT and Fg were analyzed. ResultPart one Effect of XYHT on the platelet count and functoin and the blood clotting system of wista rats and mice(1).After intervention with XYHT on normal mice,BT was decurated in a dose-dependent manner(P＜0.05).Bleeding volume of high and midst-dose groups were decreased apparently than negtive control group(P＜0.05 or P＜0.01).Clotting time of high-dose,midst-dose and low-dose groups were decurtated apparently than negtive control group(P＜0.05).BT,clotting time and bleeding volume of high and midst-dose groups were decreased apparently than positive control group(P＞0.05).(2).After intervention with XYHT on norrmal vista rats,APTT of all groups(high-dose, midst-dose and low-dose) were decurated apparently than negtive control group(P＜0.01).PT of high-dose and midst-dose groups were decurated than negtive control group obviously(P＜0.01). TT of high-dose group was decurated than negtive control group apparently(P＜0.01).There were no effect of XYHT(high-dose,midst-dose and low-dose)on the platelet count and Fg of nomal vista rats,difference had no significance(P＞0.05).Platelet aggregation induced by adenosine diphosphate(ADP) of all groups(high-dose,midst-dose and low-dose) was enhanced apparently than negtive control group(P＜0.01 or P＜0.05).Platelet aggregation induced by arachidomic acid(AA) of all groups had no significance difference than negtive control group (P＞0.05).TT and PT of all groups(high-dose,midst-dose and low-dose) were decurated apparently than positive control group(P＞0.05).APTT of high-dose and low-dose groups were decurated than positive control group obviously(P＞0.05).Platelet aggregation induced by ADP of all groups(high-dose,midst-dose and low-dose) was enhanced apparently than positive control group(P＞0.05).Part two Effect of XYHT on the platelet count,functoin and the blood clotting system of blood coagulation dysfunction mice and rats model led by Warfarin(1).After intervention with XYHT on blood coagulation dysfunction mice model,clotting time of high-dose,midst-dose and low-dose groups were decurtated apparently than negtive control group(P＜0.05 or P＜0.01).Clotting time was decurated in a dose-dependent manner (P＜0.01).Clotting time of high-dose groups were decurtated apparently than positive control group(P＞0.05).(2).After intervention with XYHT on blood coagulation dysfunction vista rats model,APTT, PT,TT of high-dose and midst-dose groups were decurated apparently than model control group (P＜0.05),APTT and TT were decurated in a dose-dependent manner(P＜0.05 or P＜0.01).Fg of high-dose group was higher than model control group(P＜0.05),platelet aggregation induced by ADP of high-dose,midst-dose groups were enhanced apparently than model control group(P ＜0.01 or P＜0.05).Platelet aggregation induced by AA of all groups had no significance difference than model control group(P＞0.05).APTT and PT of all groups(high-dose, midst-dose and low-dose) were decurated apparently than positive control group(P＞0.05).TT of high-dose and midst-dose groups were decurated than positive control group obviously(P＞0.05). Platelet aggregation induced by ADP of high-dose and midst-dose was enhanced apparently than positive control group(P＞0.05).Conclusion1 XYHT has effect of accelerating the clotting process and promoting hemostasis,it benefits to the thrombosis and hemostasis in normal animals and blood coagulation dysfunction animals led by Warfarin.2 XYHT can activate blood coagulation factor which accelerate the clotting process and promoting hemostasis by endogenous and extrinsic coagulation pathway,it enhances the platelet aggaregation function and promoting hemostasis.3 XYHT decurates bleeding time in normal animals,decrease clotting time in blood coagulation dysfunction mice led by Warfarin,decrease TT and APTT in blood coagulation dysfunction rats led by Warfarin in a dose-dependent manner.4 The effects of XYHT to the function of hemostasis and platelet aggaregation in normal animals and blood coagulation dysfunction animals were samely as that of Yunnan white powder.
