OBJECTIVE To test the in vitro killing effect of traditional Chinesemedicines on ocular demodex in order to provide a better therapy to treat demodex blepharitis.METHODS 1. Ocular Demodex mites were taken from blepharitis patientswhose eyelashes were found to have cylindrical dandruff(CD). 2. Thirty kinds of traditional Chinese herbs were chose according to《Chinese Traditional Medicine Thesaurus》,《Countrywide Chinese Traditional Medicine Compilation》and magazines that had the anti-parasite effect. 20g of each herbs were extracted with 80% ethanol by heat reflux. The killing effect of each herb on ocular demodex was recorded as the demodex survival time (ST) which was defined as the time when the demodex movement ceased after the solution was added to. Radix Stemonae, taken by many reports as an effective mite-killing herb, was used as positive control, while sodium chloride was used as negative control. 3. Tracking method was used in in-vitro mite-killing experiment and to observe morphologic change and survival time in the course. 4. Five part isolation with petroleum ether- chloroform - acetic ether- n-butanol- distilled water by heat reflux was used to test the active portion of- Fructus Xanthii. Mite-killing experiment were conducted to the five extracts to search for the effective part.RESULTS 1. The demodex survival time of 30 kinds of traditional Chineseherbs was from 6.33 minutes to 179.93 minutes. The demodex survival time in Radix Stemonae was (30.5±3.7 minutes), that in 80% ethanol was (258.53±13.19 minutes), and sodium chloride was (47.07 +++ 6.81 hours). Among them, demodex survival time in 9 kinds of herbs were shorter than Radix Stemonae, they were Flos Caryophylli (6.33±1. 75 minutes), Fructus Xanthii (12. 47±1. 41minutes), Herba Taraxaci (14.07±1.83 minutes), Cortex Meliae(15. 13±1.30minutes), Fructrs Quisqualis (19. 80±1. 57minutes), Folium Artemisiae Argyi (23.87±1. 64 minutes), Flos Genkwa (24.93±2.05 minutes), Cortex Phellodendri (26.13±1. 96minutes), Radix Sophorae Flavescentis (27.07±2. 19 minutes). 2. The effective mite-killing portion of Fructus Xanthii was glycoside, water-solubility alkaloid et al, which was extracted by n-butanol.CONCLUSIONS 1. Flos Caryophylli, Fructus Xanthii, Herba Taraxaci,Cortex Meliae, Fructrs Quisqualis, Folium Artemisiae Argyi, Flos Genkwa, Cortex Phellodendri, Radix Sophorae Flavescentis et al have better demodex-killing effect than Radix Stemonae in vitro. 2.
