Multiple factors participate in SCC's invasion. Human pappilloma virus (HPV) has intimate relationship with cervical intraepithelial neoplasia(CIN) and SCC in recently studies, but its mechanism has not been defined yet. Many evidences confirm the relationship between cell aberrant cell cycle regulation and tumorigenesis. Cell cycle G1-S is important in period transition. Cell cycle protein D1(Cyclin D1 ) plays an important role on modulation of G1-S period transition's control. Proliferation cell nuclear antigen (Ki-67) is the most significant index for proliferation capabitity in the proliferation cell nuclear antigen. It can distinguish malignant tumor and benign tumor and forecast their prognosis. Squamous cell carcinoma of cervical cancer(SCC) is a common gynecological tumor. The invasive mechanism of Cyclin D1 and Ki-67 has not been defined in CIN and SCC . The aim of this study is to investigate expression of Cyclin D1 , Ki-67 and relationship between them and HPV in CIN and SCC and to offer a new monitoring index for invasion and prognosis of CIN and SCC.Objective: 1. To observe expression of Cyclin D1, Ki-67 and their relationship with CIN and SCC, to study their invasive mechanism in CIN and SCC , to survey whether they can be used as prognosis of CIN and SCC .2. To observe expression of Cylin D1, Ki-67 and their relationship with HPV infection, to study whether they can be used as prognosis of HPV infection.Methods:Hybird capture II was used to detect expression of HPV. Immunohistoch- emical assay was used to detect expression of Cyclin D1, Ki-67, in biopsy samples. 30 normal control group are from benign uterus,176 CIN and SCC. Statistical analyses were performed with SPSS13.0 software using Chi-square test, Spearman test and Mann-Whitney test. Statistically significant level was considered as"alpha equals 0.05"。Results:1. In total 206cases, Cyclin D1 expression was mainly detected in the nucleus and cytoplasm of cervical organizations.The expression of cyclinD1 in CINⅢwas significantly higher than that in control group but downregulate in SCC.The expression of Cyclin D1 in SCC was significantly lower than all other groups (P<0.05).2. In total 206 cases, Ki-67 expression was mainly detected in the nucleus .The expression of Ki-67 in control group, CINⅠ,CINⅡ, CINⅢ, SCC was gradually increased.The expression of Ki-67 in SCC was significantly higher than that in control group, CINⅠ, CINⅡ, CINⅢ( P<0.05). The expression of Ki-67 in CINⅢand CINⅠwas significantly higher than that in control group and CINⅡ(P<0.05).3. High risk HPV was demonstrate in 47.36%, 41.18%,21.88% 100% of CINⅠ, CINⅡ, CINⅢand SCC,There was no correlation between Cyclin D1 and HPV infection . There was good correlation between Ki-67 and HPV.Conlusion: 1. The expression of Ki-67 in control group, CINⅠ,CINⅡ, CINⅢ, SCC was gradually increased,It can be used as a monitoring index for prognosis of CIN .2. There was good correlation between Ki-67 and HPV., It can be used as a monitoring index for prognosis of CIN with HPV infection.3. There is no increasing tendency of the expression of Cyclin D1 in control group, CINⅠ, CINⅡ, CINⅢ, The expression of cyclinD1 in SCC was significantly downregulate ,It is poor as a monitoring index for prognosis of CIN .4. There was no correlation between Cyclin D1 and HPV infection . It is poor as a monitoring index for prognosis of CIN with HPV infection.
