Objective: To investigate the significance of hypoxia-indueible factor-1a(HIF-1α) and its correlation with angiogenesis in precancerosis and cervival cancer by detecting the expression of HIF-1αand heparanase(Hpa) in cervical intraepithelial neoplasia(CIN) and cervival cancer, and analyzing the association between the two factors.Methods: Immunohistochemical methods were used to detect the protein expression of HIF-1αand Hpa in paraffin-embedded specimens of 80 cervical cancer, 33CINIII, 28CIN II, 31CIN I and 20 normal cervical tissue.The correlation of the two factors with the type of cervical lesions and clinical feature of cervical cancer was evaluated by chi-square test, and the relationship of the two factors was analyzed by Spearman rankcorrelation test.Results: 1.The positive rate of expressions of HIF-1αin normal cervical tissue, CINI-II, CINIII and cervical cancer were 5.0%, 20.3%, 54.5% and 77.5%, respectively.the positive percent of HIF-1αin CINIII was significantly higher than CIN I -II and normal cervical tissues (p<0.05), and which in cervical cancer was significantly Higher than CINIII(p<0.05).2.The positive percentage of expressions of Hpa in normal cervical tissue, CIN I - II, CINIII and cervical cancer were 10.0%,35.6%,63.6% and 81.2%, respectively.The positive percent of Hpa in CIN I - II was significantly higher than normal cervical Tissues (p <0.05). The positive rate of Hpa in CINIII was significantly higher than in CIN I - II (p <0.05), and cervical cancer was significantly higher than CINIII(p<0.05).3.The expression of HIF-1αhad positive correlation with the Hpa by Spearman rank correlation test(r=.0632, p <0.05).Conclusion:The positive expression rate of protein of HIF-1αand Hpa showed tendency of ascensus with the progression of cervical leisions. Furthermore they were positive related to clinical stages and negative related to histological grades.
