Objective To study the cultivation parameters and paclitaxel treatment concentration on hepatoma carcinoma cell strain HepG2 in vitro. To investigate the effects of proliferation inhibition, cell cycles, ultrastructural changes of cells and apoptosis induction of paclitaxel loaded ultrasound contrast agents on hepatoma carcinoma cell strain HepG2.Methods HepG2 cell strain was cultured in vitro , cells growth characteristics were observed by morphology research and MTT assay;The inhibitory effects of different dosages of paclitaxel on HepG2 cells proliferation were detected by MTT assay.HepG2 cell was divided into 4 groups:blank control group,paclitaxel group,ultrasound contrast agents without drugs group,paclitaxel loaded ultrasound contrast agents group. Proliferation inhibition at different time were observed with MTT assay in different groups. Cell cycles were detected by flow cytometry(FCM), ultrastructural changes of the cells were observed by transmission electron microscope(TEM) , apoptosis induction were observed with Annexin V-FITC technology.Results During the period of HepG2 cultivation , inoculate with concentration of 1×105/ml,achieve exponential phase during 24-26 hours. Certain concentration paclitaxel could inhibit the growth of HepG2 cells. The inhibition effects were positively related with the time and concentration of drug treatment, which showed an obvious time-dose-response relationship. The effects of paclitaxel loaded ultrasound contrast agents in proliferation inhibition of HepG2 cell were strongest,it could block HepG2 cell in G2/M phases,also could induce apoptosis of HepG2 cell,apoptosis body been observed.Conclusion With concentration of 1×105/ml, HepG2 cells could be treated 24 hours after inoculating. Paclitaxel could inhibit the growth of HepG2 cells in vitro. According to the formula and reference,suitable paclitaxel dosage was 1×10-7mol/L. Paclitaxel loaded ultrasound contrast agents could inhibit the proliferation of HepG2 cell,block HepG2 cell in G2/M phases,induce apoptosis of the cell.
