Diabetes is a chronic which is harmful to people's health,it's characteristic is high blood glucose level.Long-termed high blood glucose level lead to oxidative stress increasing,and cause many chronic complications.Diabetes have become one of significant epidemic disease,Its harm inferior to cardiovascular disease and tumor.So,searching for new drugs which treat diabetes,especMly the natural drugs which treat complications of diabetes have become important task to control diabetes.Natural flavonoids which have the particular pharmacological activities and high efficiency of lowing blood glucose without toxicity and side effect become the hot of studying and inventing new antidiabetic drugs.Prinsepia Utilis Royle is fruits of Prinsepia Utilis Royle in Prinsepia Royle from Rosa Ceae.According ro report of documents,Prinsepia Utilis Royle oil can decrease blood glucose and blood lipid,but have not repot about total flavonoids from Prinsepia utilis Royle decreasing blood glucose.In order to find new resource to control diabetes,the subject studies on technology of extraction,identification and examination of content of total flavonoids from Prinsepia Utilis Royle(FPR),and find best technology of extraction of the total flavonoids from Prinsepia utilis Royle,then studied on the hypoglycemic effect of total flavonoids from prinsepia utilis royle in diabetic mice,to evaluate the effects of the total flavonoids from prinsepia utilis royle on blood glucose(GLU),total cholesterol(TC), triglyceride(TG),very low density lipoprotein cholesterol(VLDL-C),alanine aminotransferase(ALT),aspartate aminotransferase(AST),blood urea nitrogen(BUN) in diabetic mice.Studies the therapeutic efficacy to complications caused by diabetes through investigating its effects on cardiac tissue,hepatic index and hepatic tissue,pulmonary tissue,renal index and renal tissue.The main result as following:1.Studies on technology of extraction of total flavonoids from Prinsepia Utilis Royle were studied with the methods of Organic solvent extraction law,ultraflltration and recrystallization.Color reaction and thin-layer chromatography were used for identification and content of flavonoids was determined by ultraviolet spectrophotometry compared with Rutin.The result showed that the methods of recrystallization is best to compared Organic solvent extraction law and ultrafiltration,the content of total flavonoids from Prinsepia Utilis RoyIe is 33.6%.2.Alloxan be injected to the mice at the dosage of 200mg/kg by the method of intraperitoneal injection.It be The classified into diabetes mellitus(DM) model group when the concentration of blood glucose exceed 10.00 mmol/L.The alloxan-induced diabetic mice were randomly divided into model control group,acarbose therapeutic group and the total flavonoids from Prinsepia utiIis RoyIe therapeutic group,acarbose therapeutic group and the total flavonoids from Prinsepia utiIis Royle therapeutic group were given acarbose (20 mg/kg)and the total flavonoids from prinsepia utilis royle(300 mg/kg)once a day by the method of intragastrical administration,model control group and normal control group were given normal saline once a day by the method of intragastrical administration.The level of GLU,TC,TG,VLDL-C,ALT,AST and BUN were detected and recorded respectively after two and four week of treatment.The result as following:compared with the same time model control group,total flavonoids from Prinsepia utilis Royle can reduced significantly the level of blood glucose,triglyceride and aspartate aminotransferase in diabetic mice(P<0.01),and reduced markedly the level of blood low density lipoprotein cholesterol,alanine aminotransferase and blood urea nitrogen in diabetic mice compared with model control group(P<0.05).3.To study the therapeutic effect of total flavonoids from Prinsepia Utilis Royle to the complications cansed by diabetes.The pathologic changes of the cardiac tissue,hepatic tissue,pulmonary tissue and renal tissue were observed under light microscope after two and four weeks treatment respectively;At fourth week,some renal tissue fixed with 2.5% glutaral in four centi-degree and their ultrastructure were observed under eletron microscope.The result as following:compared with the same time model control group. the pathological changes of each organ relieved in the total flavonoids from Prinsepia utilis Royle therapeutic group,under light microscope,longitudinal striation and sransverse striation of cardiac muscle fbers were clear relatively,cardiac muscle fibers which developed cytoplasmic eosinophilia reduced;hepatic index reduced markedly,liver cell cordc was clear,hepatic cells developed granular degeneration slightly;alveolar space augmented,alverolar walls almost recovered normoal,little reticular fiber and collagen fiber was in interstitium of pulmonary alveolus and in the surrounding of bronchi, pulmonary hemorrhage relieved;renal index reduced significantly(P<0.01),glomerular volume reduced,mesangial cells decreased,the glomerular basement membrane did not thick obviously,little fibroblast cells developed proliferation in the interstitium of renal tubule,tubular epithelial cells almost recovered normal,glucogen reduced in renal tubule, reticular fiber and collagen fiber decreased in the interstitium of renal tubule.Under eletron microscope,mitochondrions of micovillus almost recovered normal in tubular epithelial cells,foot process of podocytes lined up in order.Thus,total flavonoids from Prinsepia utilis Royle can relieve the changes of pathology of cardiac muscle,hepatic tissue, pulmonary tissue and renal tissue,and inhibite hepatic index and renal index,and delay the progression of injure of cardiac muscle,liver,lung and kidney induced by diabetes mellitus.
