Effects Of Lu's Zizao Decoction On Immunopathological Changes Of SjÇ’gren's Syndrome In Mice

Object:From the point of pathology and immunology to explore the pathogenic mechanism of Sj(?)gren's syndrome(SS) with the induced animal model,and explore therapeutic mechanism of Lushi zizaotang on SS with the induced animal model. Materials and methods:C57 mice induce by syngeneic salivary gland tissue with CFA and pertussis vccine were used as Sjogren' s syndrome model.From the second day of model made,the mice of the two treated groups were orally administreated Lu's Zizao Decoction once time daily,the mice in other two groups were fed with the same volume diluted water.Mice were killed after two weeks,and their weight were measured.Weight of drinking water daily in the last week was measured too.Fcm method was used to assessed the ratio of cells in submaxillary gland.IFN-γ, IL-10,FasandFasl in submaxillary gland cells were detected with the immunohistochemistry method.After experiment,the submaxillary gland of mice were sectiored in 6 um thickness to observe the pathological changes afer fixed in formalin,dehydrated in alcohol,and embedded in paraffin.Results:The mice weight of normal group was obvious heavier compared with that of other three groups.The mice weight of the three treated groups was similar.Weight of drinking water daily in the last week was similar beween every two groups.In the pattern group and the other two treated groups,the ratio of cells in submaxillary gland was highter than those of normal group,and in the two treated groups,the ratio of cells in submaxillary gland was decreased significantly.IFN-γ,Fas and Fasl in cells of the pattern group and the two treated groups were higher than those in the normal group,but IL-10 was lower than that of normal group.Microscopically,lymphocytic infiltration and epithelium cells in catheter proliferation were seriously in pattern group.Those of treating groups were minor.Conclusions:Lu' s Zizao Decoction can inhibit the pathology damage of SS mice by regularing the proportion of lymphocytes and the expression of IFN-γ,IL-10,Fas and Fasl.
