Objective:To study the effect of intercellular adhesion molecule-1 and vascular cell adhesion molecule-1 in regenerated endothelial cell after carotid artery injury, furthermore to investigate the mechanism of vascular restenosis.Method:Forty health male Wistar rats were divided into squeezing injury groups randomly.Carotid artery of rat was exposed under general anesthesia.Manual wood splint(13 mm×5 mm×1 mm) was set on the carotid artery separately.Then the slides were squeezed paralleling to the artery for 25 min.At days 1,7,14 and 28 after injury,rats(n=10) were killed and the target vascular sections were isolated and stained with HE to study the pathology changes.Immunohistochemistry was used to detect ICAM-1 and VCAM-1 expression in target vessels and computer image analysis was used to detect the degree of expression about ICAM-1 and VCAM-1,the thickness of artery intima and tunica media and lumen area are calculated.Results:At 7 d after injury,the neointima formed and continuously thickened. At 14 d after injury,the thickness of the neointima reached their maximal(P<0.05),at 28 d after injury,the thickness was of no difference.At 7 d after injury the lumen area had no significant change(P>0.05),At 14 to 28 d after injury,the lumen area obviously decreased(P<0.05),The thickness of the tunica media at different times after squeezing injury had no significant change.At 7,14 d after injury,the expression of ICAM-1 and VCAM-1 in intima area was higher in squeezing injury group(P<0.05),At 28 d atter injury,the expression of ICAM-1 and VCAM-1 had no significant difference(P>0.05).Conclusion:Vascular endothelial regeneration play a key role in the event of endothelial recovery,but intima hyperplasia may take part in the pathogenesis of restenosis,Detecting with immunohistochemistry,the buffy particles are present on cell membrane and in cytoplasm,and they are ICAM-1 and VCAM-1.The ICAM-1 and VCAM-1 at 7 d and 14 d after squeezing injury had statistically significant difference compared with other groups(P<0.05).
