Folk medical experience showed that water extract from leaves of Glycyrrhiza inflata Batal. is pretty effective for prostatic diseases, especially chronic nonbacterial prostatitis, but the potential basis is not clear at present. Firstly,the conditions of total flavonoids were optimized and glycyrrhizic acid content, total Sugar content were determinated. Secondly,the anti-inflammatory effect of the leaves of Glycyrrhiza inflata Batal. was researched using ear edema induced by xylene and paw edema caused by carrageen. At last,based on the good anti-inflammatory effect, studied the dose dependence of the curative effect on chronic nonbacterial prostatitis using water extract from leaves of Glycyrrhiza inflata Batal. These provided some usefuL theory for new prostate medicine research.(1)The conditions of homogenate extraction of total flavonoids were optimized by orthogonal test and response surface method respectively. The results show that homogenate extraction requires less extraction time and solvent. And the former was adding 40-fold ethanol (70%v/v), extracting for 6min. The latter was adding 40.3-fold ethanol (70.2%v/v), extracting for 5min and that the extraction rate of total flavonoids is up to 2.91%. The experimental values agreed with the predicted values .The result agreed basically between the different analytical approachs. So the optimum extraction conditions was accurate and reliable. Secondly , the content of Glycyrrhizic acid (GA) in roots, overground caudex, leaflet were mensurated by HPLC, and the roots, leaflet and overground caudex were 2.94% , 0.033% and nothing respectively . At last, the content of the total sugar from leaves of Glycyrrhiza inflata Batal is 15.93%, RSD=0.55%, which was determined by phenol-suLfuric acid.(2) By using the optimum extraction conditions and determination of 2.2 , the content of total flavonoids is 2.73%, by using the same method of 2.3, 2.4, the content of Glycyrrhizic acid and total sugar from leaves of Glycyrrhiza inflata Batal.were nothing, 5.92% respectively. The anti-inflammatory effect of the leaf of Glycyrrhiza inflata Batal. was researched using ear edema induced by xylene and paw edema caused by carrageen. The result indicated that the inhibition rate of edema of water extract and the three parts separated from water extract were higher than the control group, and especially the elution agent of ethanol (50%v/v) was best. (3)The animal models of chronic nonbacterial prostatitis were made successfully by injecting 1% carrageen injection into the prostatic back. And study on the chronic nonbacterial prostatitis treater with high, middle and low dose group of water extract from leaves of Glycyrrhiza inflata Batal. . The resuLt indicated high and middle dose group were significantly superior to positive drug group,white blood cell number significantly reduced , and lecithin corpuscuLar density significantly increased . Pathologic Biopsy showed good effect for prostatitis.
