Acute pancreatitis (AP) is one of the acute abdomens, which is a common clinical disease, having many complications, high rate of mortality, and an upward trend now. Its overall mortality rate is 5% ~ 10%, and become a serious disease to human health. At present, the earlier treatment to AP, mainly use non-surgical therapy, multi-use of traditional Chinese and chemical medicine treatment to reduce complications and the rate of mortality. Most of the patients with acute pancreatitis were recovered by conservative treatment of Chinese and Western medicine. In this paper, according to"Chinese New Drug Research Guide", we set up animal models to study the treatment of compound of traditional Chinese medicine-- Qingyilidan Granula toward the experimental acute pancreatitis in rats. A preliminary study are as follows:1. The capability of Qingyilidan Granula to inhibit the pancreatic enzyme of the experimental acute pancreatitis in the rat.Using the retrograde injection of sodium bezoar to the cholangiopancreatography to set up rat model of acute pancreatitis, to study the impact of Qingyilidan particles about serum amylase and lipase of experimental acute pancreatitis rats. The results showed that the particles can be significantly decreased the activity of serum amylase and lipase, and has a dose-effect relationship.2. The capability of Qingyilidan Granula to inhibit the injury caused by peroxide of the experimental acute pancreatitis in the rat.Using the retrograde injection of sodium bezoar to the cholangiopancreatography to set up rat model of acute pancreatitis, to study the impact of Qingyilidan particles about malondialdehyde(MDA) and superoxide dismutase(SOD)in experimental acute pancreatitis rats. The results showed that the particles can be significantly decreased the content of MDA and improved the activity of SOD, and has a dose-effect relationship. It proved that the drug can effectively improve the ability of eliminating free radicals and reducing the excessive lipid oxidation, thus reducing edema and injury of the pancreas. So we know the drug can effectively protect the experimental acute pancreatitis rats caused by the sodium bezoar.3. Qingyilidan Granula have the capable of protecting the pancreatic tissue of the experimental acute pancreatitis rats.Taking pancreatic tissues of different groups of rat models, to do pathological light microscope examination, to score the specimens in accordance with the standard score of light microscope observing. The results showed that rats who suffering from acute pancreatitis, were significantly increased in the score than the normal control groups, the group who were given Qingyilidan particles 6.3g/kg and the positive control Yidanshu particles 6g/kg group,s score were dropped significantly. Although the group(Qingyilidan particles 1.5g/kg) has some role to improve, but there was no clear statistical significance compared with the model control group. It was showed that the drug could reduce pancreatic tissue,s damage on rats with acute pancreatitis obviously.4. Anti-inflammatory effects of Qingyilidan GranulaThrough the establishment of xylene-induced swelling of mouse ear model and carrageenan induced rat foot swelling model to study cholagogic anti-inflammatory effects of Qingyilidan particles. The results show that each dose Qingyilidan particles group can significantly reduce the swelling of mouse ears, there has been significant inhibition about xylene-induced swelling of mouse ears, and has a dose-effect relationship. Each dose of Qingyilidan particles have the ability to inhibit the rat paw swelling obviously caused by carrageenan in the later 1~5 h, and as the dose increased, role of intensity enhanced, the role of the time extended. The treatment group compared with the model group had no significant difference after 6h inflammation occurred.5. Analgesic effect of Qingyilidan Granula"Acetic acid writhing" and "hot plate" method were used to study the analgesic effect of Qingyilidan particles. The results showed that Qingyilidan particles of all dose group can significantly lessen mouse writhing number, and inhibit writhing response caused by acetic acid. Although low-dose group has a certain inhibition effect, but there is no statistical significance compared with the model group; different dose group Qingyilidan particles after the administration 1h can significantly extend the mouse,s pain reaction time caused by heat stimulation and can be sustained for more than 4h.Conclusion: For acute experimental rats, Qingyilidan particles can reduce the activity of serum amylase and lipase, and improve the ability of eliminating free radicals and reduce the excessive lipid oxidation, thus reducing edema and injury of the pancreas. So we know the drug can effectively protect the experimental acute pancreatitis rats caused by the sodium bezoar. It can significantly inhibit the xylene-induced swelling of mouse ears, and has a dose-effect relationship. Qingyilidan particles could inhibit rat paw swelling caused by carrageenan, and as the dose increased, the role of intensity enhanced, the role of the time extended. It also can significantly inhibit acetic acid-induced writhing response in mice, Qingyilidan particles can significantly extend the mouse,s pain reaction time caused by heat stimulation.
