[Objective]:Candida albicans insolubleβ-glucan(CABIG) is a kind of biological response modifiers(BRM) which has extensive biological activities,such as anti-tumor activity,increasing the number of leukocytes,anti-virus activity and so on. Previous experiments of our research team isolated successfully CAIBG from the cell walls of Candida albicans,and confirmed that CAIBG was able to elevate the number of leukocyte of peripheral blood in mice and can induce the introduction of TNF-αin blood,and studied the mechanism of them in the molecular level.Further more,previous experiment in vitro confirmed that CAIBG can induce TNF-αsecretion of the human peripheral blood mononuclear cells,and had different inhibitory effects to the different tumor cells,such as malignant melanoma B16 cells, gastric carcinoma SCG-7902 cells,and pulmonary adenocarcinoma XW-05 cells.In this study,we investigated the anti-tumor activity of CAIBG in S180 tumor-bearing mice and the effect of survival to S180 tumor-bearing mice.[Method]:Firstly we extract insolubleβ-glucan from Candida albicans.In the first anti-tumor experiment,60 ICR mice were put in S180 cells,and then randomly divided into 6 groups:negative control group(NS10ml/kg·d-1),dissolvent control group(1%tween 80 10ml/kg·d-1),positive control group(CTX30mg/10ml/kg·d-1), experimental group(CAIBG100mg/10ml/kg·d-1),drug combination control group (CTX30mg/10ml/kg·o bid-1+ CAIBG100mg/10ml/kg·o bid-1),and coordination group (Bozhi Glycopeptide 20mg/10ml/kg·d-1).The difference between the twice anti-tumor experiments were the dose of drug combination control group,in the second experiment,the dose of drug combination control group were CTX20mg/10ml/kg·d-1+ CAIBG100mg/10ml/kgd-1.All the mice had intraperitoneal injection for 10 days,and at the 12th day,the weight of all the mice were measured after being killed,the tumor were dissected,the index of spleen and thymus were computed and histopathological examination were evaluated.In the first survival experiment,25 ICR mice were put in S180 cells,and then randomly divided into 5 groups:negative control group(NS10ml/kg·d-1),dissolvent control group(1%tween 80 10ml/kg·d-1),positive control group(CTX30mg/10ml/kg·d-1),experimental group (CAIBG100mg/10ml/kg·d-1),drug combination control group(CTX30mg/10ml/kg·o bid-1+ CAIBG100mg/10ml/kg·o bid-1).All the mice had intraperitoneal injection for 10 days,and then,we observe and record the life time of every mouse.In the second survival experiment,60 ICR mice were put in S180 cells,and then randomly divided into 6 groups:negative control group(NS10ml/kg·d-1),positive control group (CTX30mg/10ml/kg·d-1),experimental group(CAIBG50,100,150mg/10ml/kg·d-1), drug combination control group(CTX20mg/10ml/kgd-1+ CAIBG100mg/10ml/kg·d-1). All the mice had intraperitoneal injection for 16 days,and then,we observe and record the life time of every mouse.[Results]:The weight of tumor in mice were highly diminished in experimental group, positive control group,drug combination control group and coordination group compared with negative control group and dissolvent control group(P<0.05);and the index of spleen and thymus in mice were highly exceeded in experimental group,drug combination control group,coordination group,negative control group and dissolvent control group compared with positive control group(P<0.01).In the first survival experiment,the average life time in mice who had 10 days drugs were all longer in experimental group,positive control group,drug combination control group and coordination group compared with negative control group and dissolvent control group(P<0.05).As same as the first survival experiment,the average life time in mice who had 16 days drugs were all longer in experimental group,positive control group,drug combination control group and coordination group compared with negative control group and dissolvent control group(P<0.01).[Conclusions]:CAIBG had the effect of restraining the mouse's S180 sarcoma and can extend the life time of tumor-beating mice.Using combined with CTX,CAIBG can not only have the effect of restraining the mouse's S180 sarcoma,but also can resist the immunosuppression caused by CTX.Accordingly,CAIBG is a kind of promising biological response modifier which is worthy for further research and development.
