| Purpose: Candida albicans β-glucan (CABG) has extensive biological activity. Previous experiments have confirmed that CABG can elevate leucocytes, but its mechanism is not fully understood. In this present study, we extracted Candida albicans insoluble β-glucan (CAIBG)from cultured Candida albicans to observe whether elevated IL-1β, TNF-a can be induced in mice serum and the relationship between IL-1β, TNF-a and CAIBG increasing peripheral leucocyte.Methods: Through preliminary experiment, we found that the level of IL-1β and TNF-a reached peak 90 minutes after administration of extracted CAIBG made by ourselvies.140 male kunming mice were divided into 4 groups at random: untreated normal control, negative control, positive control and experiment group. Mice of each group except the untreated normal control group were administered cyclophosphamide intraperitoneally(100 mg/kg·d-1 for three days) to develop leucopenia model. From the 4th day of the experiment, mice of negative control group were administered sterile water for Injection intraperitoneally. For positive control and experiment group, rhG-CSF(30μg/kg·d-1) and CAIBG(51.2 mg/kg·d-1) were injected respectively on each mouse in the same way as negative control group. Continous administration intraperitoneally, once a day; Blood of mouse in each group was taken 90 minutes after administration of drug at the 1, 2, 3, 4, 5, 6, 7d respectively. All the blood samples were stored in the 4℃ refrigerator for 4 hours, and then centrifuged to get the blood serum which would be stored in the -20℃ freezer. IL-1β and TNF-a were detected by ELISA KIT after all the blood sample were collected. Peripheral blood leucocytes of all mice were counted before experiment, 1d, 3d after leucopenia model and before sacrificed. After sacrificed of the animals, spleen and thymus gland index was counted and histopathological examination was performed also. |
