Objective To investigate the expression of PAR1(protease-activated receptor-1)in nasopharyngeal carcinoma(NPC)and its significances.Methods Immunohistochemical (SP, Streptavidin-biotin peroxidase method) was used to detect PAR1 expression in 28 cases of benign nasopharyngeal tissues and 61 cases of NPC, VEGF ( vascular endothelial growth factor ) and MMP1(matrix metalloprotease-1) expression were also examined in 61 cases of NPC. We investigated the correlations between PAR1 expression and VEGF expression in 61 cases of NPC, so did the correlation between PAR1 expression and MMP1 expression. The relationship between the expression of PAR1 in NPC patients and their clinical parameters was retrospectively analyzed. PAR1 expression on protein level and mRNA level were evaluated in two human NPC cell lines (CNE-2 and CNE-1) by Western blot analysis and RT-PCR.Results Of 61 cases of NPC examined, PAR1 expressing is positive in 48 cases; of 28 cases of nasopharyngeal benign lesions examined, PAR1 expressed in 10 cases. The ratio of PAR1 expressed in NPC was much higher than that in the nasopharyngeal benign lesions (P<0.01). of 61 NPC cases'biopsy by immunohistochemistry detection, PAR1,VEGF and MMP1 positive expressing ratios were 78.7%(48/61),59.0%(41/61),77.0%(47/61) correspondingly。The positive correlation was found between PAR1 expression and VEGF expression(r=0.539,P=0.001), while such correlation was not found between PAR1 expression and MMP1 expression(r=0.186,P=0.052).The more malignant and metastasis cell line(CNE-2)expressed higher mRNA and protein level of PAR1 than the lowly malignant and metastasis cell line (CNE-1)by RT-PCR and Western blot analysis. The expression of PAR1 was correlated with patients'clinical datas such as invasive growth patterns of cancer,higher TNM clinical stags,region lymph node metastasis of NPC(P<0.05),while was not correlated with patients'ages, patients'different genders, primary tumor stages(P>0.05).Conclusions The expression of PAR1 is correlated with carcinogenesis,invasive growth patterns,more advanced malignant degree,more advanced TNM clinical stages and so on. PAR1 may contribute to the malignant phenotype and biological behaviors in NPC.
