Study on the correlation of expression between Metastasis associated-1(MTA1) and vascular endothelial growth factor(VEGF) and proliferating cell nuclear antigen(PCNA) in human Nasopharyngeal CarcinomaObjective:To study the correlation of the expression between Metastasis associated-1(MTA1) and vascular endothelial growth factor (VEGF) and proliferating cell nuclear antigen (PCNA) in different clinic grade human Nasopharyngeal Carcinoma, and discuss its significance combined pertinent literature.Methods:The expression levels of Metastasis associated-1(MTA1), VEGF and PCNA in75cases of human Nasopharyngeal Carcinoma and in10cases of normal pharyngeal mucosa tissues were detected with immunohistochemical technique and image analysis system, and the correlation of the expression of between Metastasis associated-1(MTA1) and VEGF and PCNA were analysized.Results:1. The expression of Metastasis associated-1(MTA1), VEGF and PCNA in normal pharyngeal mucosa tissues was negative.2. The expression of Metastasis associated-1(MTA1), VEGF and PCNA in human Nasopharyngeal Carcinoma was positive, and the differences of their expressin levels in different clinic grades Nasopharyngeal Carcinoma were statistically significant (P<0.05).3. The correlation of expression between Metastasis associated-1(MTA1) and VEGF and PCNA in human Nasopharyngeal Carcinoma was positive (P<0.05).Conclusion:Metastasis associated-1(MTA1) may play an important role in the main malignant biological behaviors of human Nasopharyngeal Carcinoma, which may be a potential target for human malignant Nasopharyngeal Carcinoma therapies. Relation between Expression of the MTA1gene and the property of Invasion and Metastasis in Human nasopharyngeal carcinoma CellsObjective:Nasopharyngeal carcinoma has high ability to invasive and metastasis. Tumor metastasis related gene Metastasis associated-1(MTA1) is a recently discovered tumor mestastasis candidate gene. This research is to compare the difference expression level of MTA1between the highly and lowly metastatic potent sublines(6-10B,5-8F) of nasopharyngeal carcinoma celllines, and to estimate the correlation of expression level of MTA1-EGFP with in vitro invasion potention in nasopharyngeal carcinoma cells.Method:The MTA1expression level of two cell lines was checked by semi-quantitative RT-PCR, cell invasion assay was used to evaluate the invasive capacity of two carcinoma cell lines; The lowly metastasis line (6-10B) were transfected with MTA1-EGFP full-length cDNA expression plasmid were transfected into The lowly metastasis line (6-10B) by lipofectamine; the changes of MTA1expression and invasion potential were gauged after transfection.Result:The Highly pontential metastasis cell line5-8F expressed significantly higher level of MTA1than that of lowly metastasis cellline6-10B. The invasive potential of lowly metastasis ability cell line was increased after MTA1gene transfection.Conclusion:There may be a correlation between MTA1and metastasis potential of human nasopharyngeal carcinoma cell, and the mechanism of MTA1in nasopharyngeal carcinoma metastasis and its possible role in associated gene therapy are deserved further studies. Effects of MAT1antisense oligodeoxynucleotide on expression in human nasopharyngeal carcinoma cell line CNE1and ability of invasion and metastasisObjective:To study the effect of Metastasis-associated-1phosporothioated antisense oligodeoxynucleotide on MAT-1expression and the ability of invasion and metastasis in human nasopharyngeal carcinoma cell line CNE1.Methods:The antisense, sense and nonsense oligodeoxynucleotides for Mtal gene were synthesized and delivered into the human nasopharyngeal carcinoma cell line CNE1. The effects of oligodeoxynucleotides were examined with immunohisto-chemistry, RT-PCR and Western blot.The ability of invasion and matastasis were detected by Boyden chamber invasive models.Results:The expression of MTA1was remarkably decreased after antisense oligodeoxynucleotide treated cells compared with the control cells, So as to the ability of invasion and metastasis.Conclusion:There may be a relationship between expression of MTA1and invasive potential of human nasopharyngeal carcinoma cell. The MTA1gene antisense oligodeoxynucleotide maybe can significantly inhibit the expression of MTA1and ability of invasion and metastasis in CNE1cell。...
