Clinical Relationship Between Expression Of PTEN,MMP-2 And TIMP-3 In Gastric Cardia Carcinoma

[Objective] To investigate the expression of PTEN,MMP-2 and TIMP-3 in gastric cardia carcinoma and normal gastric mucosa, and to explore their relationships with the invasion,metastasis,biological behavior and prognosis of gastric cardia carcinoma.[Methods] 115 cases of gastric cardia carcinoma surgical specimens, 40 cases of paraneoplastic gastric noncancerous tissues (the distance to cancer border>5cm) are normal control group. The expression of PTEN,MMP-2,TIMP-3 and proliferating cell nuclear antigen Ki-67 were detected by immunohistochemical method. The relationship of PTEN,MMP-2 and TIMP-3 between clinicopathological characteristics was analyzed; And the relationship with prognosis was analyzed by Kaplan-Meier life table method; Independent prognostic indicators of gastric cardia carcinoma were analyzed by COX regression model.[Results]1.The positive expression of PTEN was 61.7 % in carcinoma of the gastric cardia and 97.5% in the incisal edge of normal tissues ;The expression of PTEN was not related to the ages,sex,the degree of cancer differentiation,the size of tumor,invasion depth of tumor; but it was negatively correlated with lymphatic metastasis and stage of tumor; And it was positively correlated with the prognosis of gastric cardia carcinoma.2. The positive expression of MMP-2 was 57.4% in carcinoma of the gastric cardia and 40.0% in the incisal edge of normal tissues ; The expression of MMP-2 was not related to the ages,sex,the degree of cancer differentiation of tumor (P>0.05),but it was positively correlated with the size of tumor,invasion depth, lymphatic metastasis and the stage of tumor (P<0.05) ; And it was negatively correlated with the prognosis of gastric cardia carcinoma.3. The positive expression of TIMP-3 was 36.0% in carcinoma of the gastric cardia and 95.0% in the incisal edge of normal tissues ; The expression of TIMP-3 was not related to the ages,sex,the degree of cancer differentiation of tumor (P>0.05),but it was negatively correlated with the size of tumor,invasion depth, lymphatic metastasis and the stage of tumor (P<0.05) ; And it was positively correlated with the prognosis of gastric cardia carcinoma.4.COX regression model showed that PTEN,MMP-2,TIMP-3 and the stage of tumor were both independent prognostic indicators for survival time after operation. [Conclusion] The deletion of PTEN,TIMP-3 and the over expression of MMP-2, which may play an important role in the invasion and metastasis of gastric cardia carcinoma. Combined detecte PTEN,MMP-2 and TIMP-3 may reflect the differentiation and invasive potential of gastric cardia carcinoma tissues and it may be helpful in judging the prognosis of patients.
