ObjectiveOn the basis of comprehensive therapy with acupuncture for apoplexy at the present time,combining modern pharmacological research to prepare collagen tetramethylpyrazine sustained- elease dose,to research the effect of embedding medicine line in Baihui and Dazhui acupoints on Urokinase-type plasminogen activator(uPA) and its inhibitors-1(PAI-1) and The protective effect of Cerebral microvasculature after focal cerebral ischemia-reperfusion.MethodsChoose 112 healthy SD rats were randomly divided into six groups, 7 normal group,sham-operated group,21(1d,3d,5d group,n = 7), model group,21(1d,3d,5d group,n= 7),acupoint injection group, 21(1d,3d,5d group,n = 7),tail vein injection group,21(1d, 3d,5d group,n=7),embedding medicine line in acupoints group,21 (1d,3d,5d group,n= 7).The use of middle cerebral artery suture (MCAO) ischemia-reperfusion model,then embedding medicine in Baihui and Dazhui acupoints with collagen tetramethylpyrazine sustained-release dose.The group phase of the six rats were taken to 10%chloral hydrate anesthesia,cardiac perfusion,rapid access to complete irrigation brain cortex organizations take part in the liquid at a fixed and paraffin-embedded,the use of immunohis tochemical Detection of ischemic cortex uPA,PAI-1 content,as well as drug line points buried the impact of these indicators,and the remaining rats in each group in the broken phase directly from the fresh part of the brain cortex,in the electron microscope observation of ischemic cortex of the brain microvascular ultrastructure;and the effect of all the above index of embedding medicine line in acupoints were observed.The entire datum was analyzed by statistics.Results1.The expression of uPA heightened and the expression of PAI-1 depressed in brain tissue after focal cerebral ischemiareperfusion with the method of immunohistochemistry.The point-injection and tail vein injection compared with model group, Treatment groups can down-regulate uPA expression and up-regulate PAI-1' expression,the point-injection group and tail vein injection group compared with model group(P<0.01),but the regulation effect of uPA and PAI-1 was not clear with embedding medicine line in acupoints ld group(compared with model group,P>0.05).The preponderant effect of embedding medicine line in acupoints group represented at 3d and 5d,compared with model group(P<0.01);compared with point-injection group and tail vein injection group(P<0.05);Better than the point-injection group and tail vein injection group.2.Electron normally cessation between implant and sham rats cortical brain microvascular structure is normal,the lumen size is normal,endothelial cells and vascular smooth without the swelling,the basement membrane continuous complete.The model groups show ischemia area surrounding edema,and microvascular endothelialcell apoptosis,luminal stenosis,siltation,basement membrane thickness continuity,partial thickening,or dissolution, fracture,defect,capillary happen become pyknotic change; point-injection,the tail vein injection and the acupoint embedding medicine line is a microscopic quad model group got obvious improvement,Capillaries surrounding edema model group than that of light,basement membrane basic normal,endothelial ceils is already close to normal,embedding medicine line in acupoints group to improve the most significant,is superior to point-injection group and the tail vein injection group.ConclusionThe expression of uPA increased and the expression of PAI -1 reduced in ischemical brain tissue after focal cerebral ischemia reperfusion in rats,which reduce cerebral microvasculature basement membrane and EC serious damage,cause brain microvascular morphological structure of incomplete,aggravate ischemic reperfusion injury.Embedding medicine line in acupoints can decrease uPh expression and increase PAI -1' expression,so it can reduce cerebral microvasculature of basement membrane and EC's damage,protect the structure' integrality of cerebral microvas, and bring into play the effect of cerebral protection consequently; Its role is better than point-injection group and the tail vein injection group.
