After the Pharmacodynamics study that Hypericum ascyron L.has an evident anti-depression effect,HongHanLian granule for anti-depression is composed of active components from the extracts with appropriate adjuvants.Under the guidance of the theories of TCM and Pharmacy,the granule has been studied on pre-clinical pharmacy,which mainly includes the active components screening,quality standard,preparation procedure.Classic antidepressant models were used to primaryly screen active components ,and the result was that 30%ethanol elution show antidepressant activity in the forced swimming test in mice and tail-suspension test in mice. Flavonoids were identified in the systematic pretesting.According to the physical and chemical properties of active components of the Herb,We adopted Box-Behnken response surface methodology(RSM) to select the optimized alcohol-extract technology by using total flavone as assessing norm.And polyamide column chromatography was used to separate the active components;the study was performanced to optimize the preparation procedure with the formation,hygroscopicity,dissolubility as the synthetic criteria.In order to control the quality of active components and guarantee the clinical effectiveness,the thin layer chromatographymethod(TLC),Ultraviolet-visible spectrophotometry(UV),high performance liquid chromatography method(HPLC) were established by which the contents of total flavone and Hyperoside were determined.The research result indicates that 30%ethanol elution is active components ,the standard is controllable,optional preparation procedure is convenient and feasible as well.
