Objective Star3 signaling transduction pathway plays an important role in tumorigenesis and tumor progression.To make clear the role which Stat3 signaling transduction pathway played in tumorigenesis,invasion of esophageal squamous cell carcinoma(ESCC),we intended to detect the expression of Stat3,CyclinD1 and Bcl-2 in a ESCC tissue microarray(TMA).We hope to supply a theoretical evidence and significant marker for early diagnosis,judgment of prognosis and tumor therapy.Methods We applied tissue microarray technique and immunohistochemistry(IHC) method to detect Stat3,CyclinD1 and Bcl-2 protein expressions and analyzed their relationship with clinical data in a ESCC tissue microarray containing 860 specimens of ESCC,198 specimens of dysplasia,201 specimens basal cell hyperplasia and 467 specimens of normal mucosa adjacent to cancer tissue.All data were processed by SPSS version 13.0 analysis software.Results 1.Stat3,CyclinD1 and Bcl-2 protein were respectively detected in ESCC and dysplasia, which were significantly higher than those of control group(p<0.05).2.The expression of Star3,Bcl-2 in different histological types of ESCC were various.Middle-low grade>high grade,and the differences were significant(p<0.05);there was significant difference between the expression of CyclinD1 in high grade and low grade ESCC.3.The positive ratio of the expression on Stat3,CyclinD1 and Bcl-2 were significantly related to lymph node metastasis(p<0.05),but not correlated with the age,gender,primary location,the size of tumor and gross appearance type(p>0.05).4.There was significantly positive association between Stat3 expression and CyclinD1 expression(r=0.699,p=0.000).The expression of Stat3 was positive related to the Bcl-2 expression(r=0.660,p=0.000).Conclusion 1.The increased expression of Star3 protein in ESCC suggested it involved in carcinogenesis of ESCC.We detected that the expression of Stat3 was much higher with lymph node metastasis.It proved that Stat3 protein had a relationship with metastasis function in ESCC.2.The expression of Stat3 in primary ESCC is gradually higher following the decreased of tumor cell differentiated.This suggested Stat3 played an important role in proliferation of tumor cells.3.The expression of Stat3 was significantly associated with the expression of CyclinD1,which suggested there maybe exist cooperation between them.There were significant correlation between Stat3 and Bcl-2,which showed Stat3 could up-regulate the expression of Bcl-2.There was no significant relationship bewteen CyclinD1 and Bcl-2, which suggested they might have different mechanism on depressing the apoptosis of tumor cells and inducing tumor angiogenesis.They played different roles in carcinogenesis and growth of human ESCC.
