Objective To explore multiplication effect of PKC-δon the human hepatoma carcinoma cell.Methods Experiment in vitro:Culturing SMMC-7721 in vitro,amplification with objection gene segment,gene transduction in vitro by liposome,tansducting liposomes genes out of body,we reseached the effect of PKC-δto SMMC-7721 through the methods and techniques such as cell counting,cell morphology observering,the method of MTT,the technique of flow cytometry; Experiment in vivo:inject SMMC-7721 cell to the back of nude mice,the back of nude mice outgrow tumor,then divide into trio,inject physiological saline,pcDNA3,PKC-δseperaetly,inject one time by two days,record tumor size and take photos every time,after injecting medicine fiften times,put nude mice to death,to get the back tumor of the nude mice,make pathological section and do HE dyeing,Immunohistochemistry,Results Experiment in vitro:The methods of cell counting cell growth curved shape displayed that SMMC-7721 gradully discreased by leading in PKC-δ,the method of MTT displayed that SMMC-7721 OD dercreased,living cell remarkably decreased(p<0,01)by adding in PKC-δ,cell morphology observering displayed that cell abscisic,link loosen,lose glare by leading in PKC-δ;and the technique of flow cytometry displayed that SMMC-7721 apotosis rate creased by leading in PKC-δ;Experiment in vivo:the back tumor of the nude mice gradutely reduced by leading in PKC-δ,but the other two groups' tumor changed very small,HE dyeing can tell us that the SMMC-7721cell leaded in PKC-δpycnosis,round,cell nucleus and cytoplast dyeing densely,cell nucleus graininess,ring-shaped or crescent,cytoplast see "swollen"phenomenon and so on,those is changed in morphological apotosis.Immunohistochemistry display that SMMC-7721 cell nucleolus chang blue,cytolmph chang claybank by leading PKC-δ.Conelusions PKC-δcould inhabit SMMC-7721 to grow and promote its apoptosis.
