Effects Of Lard And Rapeseed Oil On The Formation Of Colon Cancer And The Serum Glucose And Lipid Levels Of Mice

Objective:To observe the effects of our traditional lard and rapeseed oil on the AOM-induced colon tumors in mice,And to explore with colon tumors the relationship between the fecal pH and the fecal bile acid.Meanwhile,to studies the influence of these two oils on blood glucose(BG),serum total cholesterol(TC) and serum triglyceride(TG).Methods:Mice were randomly divided into 4 groups,Control Group(CG),AOM control group(ACG),Rapeseed-oil Group(RG) and Lard-oil Group(LG).All mice were fed rodent chow respectively.To induce mice colon cancer,ACG,RG and LG received intraperitoneal injections of AOM at the 7-day,the 14-day,21-day,28-day.During the experiment,the feces pH was examined every two weeks,feces cholic acid was done every four weeks.After the mice were sacrificed,the blood from mice eyeballs were taken to measure blood glucose,serum total cholesterol and serum triglyceride level; the internal organs were taken for calculating their organ coefficients;the colon were taken to count the number of colon tumor and ACF.Results:1) The feces pH has no significant differences between the male control group of RG and ACG(P>0.05),but the feces pH in the female control group of RG was significantly higher than ACG(P<0.05).Both of male and female mice fecal pH in LG were significantly higher than that in ACG and RG(P<0.05)2) No significant difference was found in feces choice acid between all groups (P>0.05).No gender differences.3) Mice in each group have low incidence of colon neoplasms,and no significant difference was found(P>0.05).No gender differences.4) Comparison of the overall and male,no significant difference was found in the ratio of heart weight to body weight(H/B) and the ratio of kidney to body weight (K/B) between all groups(P>0.05).The ratio of liver to body weight(L/B) and the ratio of spleen to body weight(S/B) were increased in ACG than CG(P<0.05). The ratio of testicle to body weight in RG was significantly lower than in CG (P<0.05).5) No significant difference was found in the ratio of heart weight to body weight(H/B),the ratio of liver to body weight(L/B) and the ratio of spleen to body weight(S/B) of male control group between all groups(P>0.05).The ratio of kidney to body weight(K/B)in ACG,RG and LG was significantly lower than that in CG(P<0.05).6) No significant difference was found in blood glucose between ACG,RG and LG (P>0.05).But blood glucose in CG was significantly decreased(P<0.05).No gender differences.7) Comparison of the overall and male,no significant difference was found in serum total cholesterol between all groups(P>0.05).But in the female control group,the serum total cholesterol in RG and LG was significantly higher than that in ACG and CG(P<0.05).8) AOM can very significant increase serum triglyceride in mice.The male's serum triglyceride significantly has higher than the female's(P<0.05).9) The serum triglyceride in RG,LG and CG was significantly lower than that in ACG(P<0.05).The serum triglyceride in LG was significantly higher than that in RG(P<0.05);But in the female control group,no significantly difference was found in serum triglyceride between LG and ACG.Conclusion1) Kunming mouse is not sensitive to AOMwhich is the colon tumor-specific carcinogen.Can not be used as laboratory animals that AOM-induce mice colon cancer.2) AOM was significantly increased the blood glucose and serum triglyceride levels of mice.There are gender differences of serum triglyceride in AOM.But AOM make no impression on serum total cholesterol.This is our first exclusive,and there is no relevant reports at home and abroad.3) There is different between rapeseed oil and lard on the fecal pH.Lard was significantly increased fecal pH;But rapeseed oil was significantly decreased female's fecal pH.There is no different of fat kind and vary fat content in the fecal bile acid.4) Due to the very low incidence of colon cancer,we cannot observed the different of the incidence of colon cancer in fecal bile acid content.5) Lard was significantly increased fecal pH,and the number of ACF per mouse, especially male mouse.Therefore lard may be an important factor that promote colon cancer's incidence.6) AOM has different effects on organs.AOM can make mouse's liver and spleen hyperplasia in male,and kidney damage in female.7) Fatty cid composition and content had no effect on blood glucose in mice which caused by AOM.8) On the AOM,lard can be significantly increased female's serum total cholesterol; and it also decreased Hypertriglyceridemia which caused by AOM,especially male.9) On the AOM,rapeseed oil can be significantly increased female's serum total cholesterol;and it also decreased Hypertriglyceridemia which caused by AOM.10) Rapeseed oil and lard both can be significantly decreased Hypertriglyceridemia which caused by AOM.,especially rapeseed.
