| Rosemary is a sort of rising medicalplant,there are many components of rosemary had strong antioxidant activity(Antioxidant,Anti-virus,Anti-tumor,Antibacterial,Anti-inflammatory etc.) Rosemary antioxidant's used and developed in Foods,Oil production,Health-care,Cosmetics etc.,for its antioxidant activity.Recently,the extraction technic and application of active ingredient are concerned by whole world,which had been a new focus.Carnosic acid is a main active component of rosemary,which has been used in Health-care and Food Additives widely.As soon as the market demand has been getting larger,the demand reqires proving the extraction technic of carnosic acid.The main research contents of this paper included purification and extraction carnosic acid from rosemary,the extraction included distillation,homogenate,decolorization,column chromatography etc..Then optimization homogenate extraction by used response surface optimization software.Found the best homogenate condition for extraction carnosic acid,carnosol and rosmarinic acid all.Purification crude extracts by further column chromatography for getting more higher purity carnosic acid production.Purification we determined is followed:Homogenate Experiment was used for the material which was extracted from essential oil,the best condition is:liquid-solid ratio is 10.62:1 mL·g-1,ethanol Volume fraction are 79.71%,homogenate time is 5.17 min,frequency is10013.22r·min-1,3 times.The experiment shows that:the carnosol extraction ratio is 0.42%,purity is 4.87%;carnosic acid extraction ratio can be 1.48%,and purity is 21.09%;rosmarinic acid extraction ratio is 0.24%,purity is 4.33%.The best column chromatography condition:eluant is acetic ester:n-hexane is 4:6;amount of silica gel are 100-200;silica gel moisture content is 6%;diameter height ratio is 1:10;flow velocity is 4ml/min;0.3g crude extract:20g silica gel.The result showed that the purity of carnosic acid can be more than 93%,and the percent recovery can be 89%.
