Objective: Glioma is one of the most common tumors in the brain cancer,and it is about50%among the brain tumors. Although the surgical,chemotherapy and radiotherapy have developmentally enormously, thetreatment of glioma has not changed much, and the prognosis is still very poor.Along with the concept of brain tumor stem cells take out, it provides us a newidea to recognize the glimoa. This experiment is to explore the expression of theglioma stem cell markers: CD133、SSEA-1and Nestin in human glioma cell ofthe patients, and to see whether the expression of CD133、SSEA-1and Nestinhave some associate with the differentiation and pathologic grading. And tostudy these three markers whether or not can reveal some essential qualities ofthe glimoa and have some clinical significance in diagnosis and malignantdegree of judgment, or have provided the rationale of molecular biology totreatment.Methods:54cases of human glioma tissue were collected from the FirstAffiliated Hospital of GuangXi Medical University (September2011toDecember2012), and all the tissues are glioma that have been confirmed bypathological. According to the WHO grading standards of2007, gradeⅠ6cases,grade Ⅱ16cases,grade Ⅲ13cases and grade Ⅳ19cases. In addition, 6cases of normal human tissues have been for control group.Immunohistochemistry method was used to examine the level of CD133,SSEA-1and Nestin in surgical samples from54patients suffered from gliomaand6cases of normal brain tissue, and the correlation among the expression ofCD133, SSEA-1and Nestin and glioma grade was analyzed.Results:①The positive cell rates of the CD133expression were25.38%in54cases of human glioma tissues and0%in6cases normal tissues of controlgroup (P<0.05);SSEA-1were respectively26.62%and0%(P<0.05); Nestinwere respectively22.39%and0%(P<0.05)。②Compare the differentpathological grades with the positive cell rates of CD133、SSEA-1and Nestin.There were no significant difference between gradeⅠand gradeⅡ(P>0.05);There were significant difference between gradeⅠand grade Ⅲ or grade Ⅳ(P<0.01);There were difference between gradeⅡand grade Ⅲ or grade Ⅳ(P<0.05). There were significance difference between the grade Ⅲ and gradeⅣ (P<0.05). The higher pathological grades had higher positive cellrates.SSEA-1.③There were significant difference between the positive cell ratesof CD133group and SSEA-1or Nestin group (P<0.05). There were significantcorrelation between CD133group and SSEA-1group(Cp=0.84,P<0.05),CD133and Nestin (Cp=0.48, p<0.05),SSEA-1and Nestin (Cp=0.44,p<0.05).Conclusion:①The expression of CD133,SSEA-1or Nestin in differentpathological grading of glioma also had different degree expression.;and thereexpression was positive correlation with the pathological grading;this indicatethat these three markers’high expression had some relation with the malignancydegree of glioma.②It was useful in judging malignancy degree and diagnosing by detect CD133、SSEA-1or Nestin the expression in glioma.③CD133、SSEA-1and Nestin will be the most important glioma stem cells’ markers, andmay be significant in diagnosing, prognosis or treatment. They will act as moresubstantial role in investigation and combined therapy for individual in thefuture.
