The Expression Of PCNA And TNF-α On Liver Regeneration After Partial Hepatic Resection Under Different Hepatic Inflow Occlusion Methods In Rats

ObjectiveThis research is compared with three methods of the influence which regenerates to the liver through after three kinds of hepatic blood inflow occlusion 70% hepatectomy were induced the liver regeneration.Methods120 male SD rats,weighing 200～250g were randomly divided into hepatic blood inflow occlusion without hemihepatic artery control group(WH group),semi-hepatic vascular occlusion group(HH group) and Pringle group(PR group),each group using the corresponding methods of hepatic occlusion rats liver for 15 minutes。With bulldog clamp we resect median lobe and left internal lobe and left outer lobe(70% heaptectomy) to induce liver regeneration.Eight rats in each group were killed at 6 hours,12 hours,24 hours,4 days and 7 days after hepatectomy,respectively.The recording of pathological and morphological changes were observed first.The liver to body weight Ratio and Cells count undergoing mitosis in histological section,and at different time point is done in order to assess the liver regeneration.By detecting expression of PCNA,serum concentration of TNF-αwere measured at each time point, we can value the effect on liver regeneration were measured at each time point.Rusult1.Liver to body weight ratioAt each experimental group,with the time liver to body weight ratio gradually increase;One days the liver weight had no significant differences in body weight in each group;At 4 days and 7 days in WH group and HH group liver to body weight greater than in PR group,and liver to body weight of the WH group and the HH group were no significant differences.2.Hepatocyte Mitotic count(Index)At PR groups Hepatocyte Mitotic count gradually increased from 1 day after operation to 7 days after operation;At WH and HH groups hepatocyte Mitotic count increased at 1 day after operation,4 day hepatocyte Mitotic count reach a maximum; At 7 days after it has been reduced.At 1 day and 4 days after operation hepatocyte mitotic count in WH and HH group than those in PR group,while at WH group and HH group hepatocyte Mitotic count is no significant differences;At 7 days after operation, there are no difference among the groups3.Expression of PCNAEach group of expression of PCNA after the operation at 6h group has not differentiated with the normal group;at 12h after the operation,in WH group and HH group expression of PCNA increases;at 24h after the operation each group of expression of PCNA increases,the WH group and the HH group have the difference with the PR group.Among each time points the difference of the WH group and the HH group is not remarkable.4.Blood serum TNF-αlevelEach group of TNF-αlevels elevate at 6 hours after the operation,elevates by the PR group obviously,TNF-αalready gradually reduced at 12 hour after the operation in each group.Conclusion1.In the WH group and the HH group,PCNA and TNF-αhave no significant difference because the two groups may be no significant difference in liver regeneration.2.In the WH group and PR group,PCNA and TNF-αhave significant differences, because the two groups may be significant difference in liver regeneration.
