Objective:The warm water extraction and ethanol precipitation.was employed to the high molecule saccharides and micromolecule saccharide from liquid submerged fermentation extract of H.caput-medusae (Bull.:Fr.) Pers,and research on the pharmacology activity of anti-tumor,raising the counts of white blood cell, antioxidation and anti - bacterium,to establish the foundation of comprehend and exploitation more reasonable of the special precious eumycete resources—H.caput-medusae(Bull.:Fr.) Pers.Methods:1 In vitro,three tumor cells(SMMC7721,SPC-a-1,SGC7901) were cultured with HFP and HFA at the final concentrations(2000,1000,500,250,125,62.5,31.25,15.625μg/ml) following incubation,MTT chromatometry was used to measure the effect of HFP and HFA on the activity of tumor cells.2.In vivo experimentmouse portability sarcoma 180 solid tumor model was established to observe the antineoplastic activity of HFP and its effects of auxodrome,the index of spleen and thymus.3.The methods including Fe(KCN)3-TCA method,pyrogallic acid method and Fe2+/H2O2 method were used to study the effect of HFP and HFA on the reducing power,cleaning superoxide anion free radicle,cleaning hydroxy free radicle in vitro.4.Mice model of leukopenia was established to observe the rising peripheral white blood cells activities of HFP and its effects bone marrow nucleated cells,the coefficient of the spleen and thymus.5.To separate pathogenic bacterium from diarrhea mouse,and observe the anti-bacterium activities of HFP.Results:1.In vitro anti-tumor experiment:HFP and HFA both have the anti-tumor activies,when they were treated with three cancer cell lines(SMMC7721,SPC-a-1,SGC7901),but its activies were relate with cell category,administration dosage and administration time.The suppressive effection to SPC-a-1 is stronger than SGC7901 and SMMC7721,high administration dosage is better than low,and long administration time is better too.2.In vivo anti-tumor experiment:HFP may inhibited the growth of S180 tumor in mice.And the effects of the prevention group is higher than the treatment group,the high dose is better than low-dose group.And the HFP group's spleen and thymus coefficient are higher than the CTX group obviously.3.HFP and HFA have certain activeness of antioxygen,and with the same density,the effects of HFP is better than HFA.4.HFP could improve the mice modle of leucopenia,and may alleviate the degression of the number of bone marrow nucleated cell and the coefficient of spleen and thymus.But the effect of HFP independent is not better than the synergism effect of different origins polysaccharides. 5.The result of the turbidimetric method is confirmed that HFP may inhibit the grow of E.coli in vitro,and the result of MIC texting is 40mg/ml.The result is showed that liquid submerged fermentation extract of H.caput-medusae(Bull.:Fr.) Pers has the widespread Pharmacology activeness,its effective component is the saccharides which the high molecule is better than micromolecule.
