H.caput-medusae (Bun.:Fr) Pers is in rich of nutrition, it is a kind of edible and drug fungi. It contains generous protein, crude fat, ash, necessitous mineral element, saccharides, organic acid, alkaloid series, steroids and various kinds of amine acid. As the characteristic fungi resources of Mt. Changbai Nature Reserve Area, there are upstanding pharmacology effects in H.caput-medusa (Bun.:Fr) Pers. But the study concentrates upon gastrelcoma and stomach ailment, while there is less studies on polysaccharides of H.caput-medusae (Bun.:Fr) Pers. This study is aim to search for polysaccharides of H.caput-medusae (Bun.:Fr) Pers overall to offer the rationale and experimental data in application.The research uses water extracting and alcohol sinking to get polysaccharides HP1, HP2 and HP3 from H.caput-medusae (Bun.:Fr) Pers by three kinds of alcohol density. The effects on the tumor cells of the three kinds of constituents is detected by MTT method, and make sure that HP2 is the best component. The content of mineral element and polysaccharide is detected by flame aomic absorption. Then research all around liveness of HP2:(1)anti-tumor activity:study on the effects of HP2 with liveness,excretion of MMPs and cell cycle of Hela cells by the methods of MTT, gelatinase detected and flow cytometry. (2)pharmaceutic adjuvants:HP2 is combined with 5-Fu to investigate the feasibility as a pharmaceutic adjuvant using MTT and flow cytometry. (3)improve immunity:establish hypoimmunity model of mice by the drug cyclophosphamide and accordes HP2, then to detect the lymphocyte transformation and carbon clearance. (4)anti-oxidant:to set up scathing PK cells by H2O2 model and detected the saving effects of HP2. To set up hypoimmunity model of mice to detecet SOD and MDA level of mice liver and serum.Experimental results:(1)HP2 is the best content to depress the growth of Hela cells. HP2 contains 83% polysaccharide and Cu 0.01%, Pb 0%, Zn 0.07%, Fe 1.99%, Ca 10.21%, Mg 0.44%. (2)HP2 possesses the cell inhibition and dose dependent. HP2 can make MMP2 down regulation and fail to understand MMP9. HP2 can block the cell at G0/G1 phage. (3)HP2 combines with 5-Fu shows the evident effects on the Hela cell inhibition. Drug combination is better than 5-Fu alone. This hints that HP2 promotes the effects of 5-Fu. (4)HP2 can reinforce the conversion efficiency of lymphocyte, carbon clearance, thymus and spleen index whie the liver and kidney index down. (5)HP2 can repair cell damage of PK induced by H2O2, and the results with content of HP2 is direct proportion. Under the using of HP2, the level of MDA is down and SOD is up in mice.Above all, polysaccharide of H.caput-medusae (Bun.:Fr) Pers possesses favourable biologic activity and is worth to study more.
