[Objective]To investigate the expression of Cx43 and E-cad in Bladder Transitional Cell Carcinomal(BTCC) and the relationship between these expressions and clinicopathological,providing an experiment evidence for the diagnosis and treatment of the BTCC.[Medthods]The first of all , 76 cases of BTCC tissue and 22 cases of normal bladder tissue were made into the tissue-microarray, then the expression of Cx43 and E-cad in BTCC tissue and normal bladder tissue were detected by immunohistochemical S-P method,and to analyze the relationship between the expression and some clinical-pathological factors such as clinical TNM stage ,histological grade ,axillary lymph node metastasis ,recurring. and there correlation was then analyzed.[Results]1.The result of stained: In normal bladder tissue ,Cx43 and E-cad were expressed by normal epithelia with typical membranous staining,some were Cytoplasm staining; but there were mainly expressed in Cytoplasm in BTTC,Some were membranous staining.2.The positive expression rates of Cx43 in BTTC and normal bladder tissue were 42.1%,95.5%, There was significant difference with the normal bladder tissue(p=0.001);the positive expression rates in G1,G2 and G3 were 71.4%,33.3%,25.0%,the positive expression rates in Tis-T1,T2-T4 were57.8%,19.4%,which is relevant to the grading and the clinical staging of BTCC by Statistics analysis(p=0.005,0.001 respectively).3.In BTTC,the expression rates of Cx43 in the metastasized and non-metastasized lymph node groups were 54.9%,16.0%,which is closely related to the lymph node metastasis of BTTC (p=0.001).4.In BTTC,the expression rates of Cx43 in the primary tumors and recurrences groups were 38.5%,50%, The positive expressions of Cx43 between the primary tumors and recurrences were significantly different (p=0.034).5.The positive expression rates of E-cad in BTTC and normal bladder tissue were 40.8%,90.9%,There was significant difference with the normal bladder tissue(p=0.001).the positive expression rates in G1,G2 and G3 were 76.2%,30.8%,18.8%,the positive expression rates in Tis-T1,T2-T4 were 55.6%,19.4%,which is relevant to the grading and the clinical staging of BTCC(p=0.001,0.017 respectively) by Statistics analysis.6.In BTTC,the expression rates of E-cad in the metastasized and non-metastasized lymph node groups were 49.0%,24.0%,which is closely related to the lymph node metastasis of BTTC (p=0.037).7.In BTTC,the expression rates of E-cad in the primary tumors and recurrences groups were40.4%,41.7% respectively.The positive expression of Cx43 and E-cad between the primary tumors and recurrences were not significantly different (p=0.0916).8.Spearman correlation analysis revealed that the expression of Cx43 and E-cad was positively correlated in BTTC (rs=0.674; p=0.001).[Conclusions]1.This experiment confirmed that it can save time, cost of experiment and increase the efficiency if we used tissue-microarray technique combineing with immunohistochemistry technique in experience ,they can be use for bladder cancer research.2. Low expression of Cx43 and E-cad was observed in BTTC,they might be connected the occurrence,development of BTCC.The positive express of Cx43 and E-cad were associated with the lymph node metastasis and recurrence of BTTC.suggest that the expression of Cx43 and E-cadherin might be associated with the invasion and translation of tumor.3.significant correlations were found between the expression of Cx43 and E-cad in patients with BTTC,they both were associated with the mechanism of occurrence ,development, invasion,metastasis,and prognosis of BTTC. The combined detection may be contributed to the monitoring of clinical progress.
