Panpax notoginseng(Burk)F.HChen is a highly valued and important traditional Chinese herbs and belongs to the family Araliaceae. There are a wide variety of therapeutic uses of Notoginseng, including promotion of blood circulation, removal of blood stasis, induction of blood clotting, relief of swelling and alleviation of pain. Saponins and flavonoids which were reported to be the biologically active constituents are responsible for the therapeutic action of this drug.Many studies about the Saponins content and identification of Notoginseng have been reported. So far more than 60 saponins have been isolated from different parts of Notoginseng. Thus, there is no systematical study about the total flavone content and identification in flowers of Notoginseng.The orthogonal test was employed to check influences of four factors: concentration of alcohol, extraction time, frequency of extraction and ratio of liquid to material, selecting amount of flavonoids as standard according to L9(34) orthogonal design table.DM130 macroporous resin was used to separate total flavonoids in flowers of Panapx notoginseng (Burk)F. H Chen. The static adsorption capacity, the static elution ratio and the dynamic adsorption capacity were studied. The optimized conditions are as follows: solution concentration at 0.06 mg/mL, pH at 4 and the flow rate of absorption solution at 1BV/h ; using 70%(v/v) ethanol as the elution solvent, 1BV/h flow rate, within 6BV the flavonoids can be eluted completely, the adsorption ratio and elution ration can reach 71.80% and 93.37% respectively. The results showed that DM130 macroporous resin possesses high absorption and desorption capacity. It can be used to purify and separate flavonoids in flowers of Notoginseng.The ethanol extraction of flowers of Notoginseng was separated by polyamide column chromatography and recrystallization. Structures of the pure compound were established on the basis of its chemical and spectral data. A compound named kaempferol-3-O-β-D-glucopranoside is obtained and this compound is isolated from flowers of Notoginseng for the first time.
