ObjectiveTo observe the effects of herbal compound for reinforcing kidney,activating blood and arousing consciousness(HCRAA) on the ABR threshold and the expression of neurotrophic Factor 3(NT-3) of the cochlea in gentamicin(GM)-induced ototoxic mice.MethodsHerbal compound for reinforcing kidney,activating blood and arousing consciousness (prescription consists of:Moreover,Drynariae,epimedium,astragalus,Chinese angelica,Salvia,Chuanxiong,Acorus gramineus,leech,magnets,etc.) were decocted for high,medium and low concentrations of the three decoction containing respectively pharmacognostic 2.375g/ml,0.475g/ml,0.095g/ml(mouse body weight by 25g,the volume of 0.8ml orally every day, the decoction of the concentration of one is equivalent to 70kg adult day clinical routine The equivalent amount of volume,high concentration and low concentration of decoction decoction decoction drug concentration equivalent to the amount of 5 times and 0.2 times the dose).60 Kunming mice were randomly divided into 5 groups of 12.A,B,C,D group:HCRAA high,medium and low concentration of the intervention group and intervention group with normal saline by of every day, 100mg/kg,for 15 consecutive days.From the first day of GM intraperitoneal injection,four groups respectively lavage for high,medium and low concentration of Chinese compound decoction and normal saline,for 20 days;E groups:control group,normal feeding,not any treatment.Administered after the mice in each group measured auditory brainstem evoked potentials(ABR) thresholds, cochlear organizations Western blot(Western Blot) were detected in each group cochlear tissues the expression of NT-3.ResultsHCRAA of high and middle concentration could lower the elevated ABR threshold(P<0.01),and elevated the expression of NT-3 in the cochlea induced by GM(P<0.01).ConclusionHCRAA can lower the elevated ABR threshold induced by gentamycin effectively.Their mechanisms may be related to cochlear hair cell and neurone be protected by activating blood circulation and elevating the expression of NT-3 in the cochlea induced by GM.HCRAA could elevat the expression of NT-3 in the cochlea induced by GM,thereby protect the acouesthesia of gentamicin-incluced ototoxic mice.Possible that some way through the following:①Through antagonism of gentamicin on hair cell damage and preserve normal secretion of NT-3 functions; ②Certain components of Chinese medicine through the cochlear blood labyrinth barrier(BLB) to enter the cochlea,nervous and endocrine regulation,and enhance NT-3 autocrine and paracrine functions,and to promote its relations with the SGNs surface receptor binding,the maintenance of nerve cells transfer function of the normal.Studies show that physical or cochlear damage on the lower level when the neurons can listen to each other target organs,NT-3 can also be a higher level through reverse transmission neurons to SGNs;③Compound Chinese medicine for the whole theory of governance,the focus on overall conditioning,cochlear with increased NT-3 content may also be conditioned on a partial general performance of NT-3 is a small molecule protein(molecular weight: 14kd),can be thoroughly have cochlear blood labyrinth barrier and the blood-brain barrier,which also can enhance the body's overall NT-3 level,by way of blood circulation or cerebrospinal fluid within the cochlea arrival.I think Chinese medicine HCRAA protects against gentamicin ototoxicity reaction mechanism,in addition to improving the systemic and cochlear blood circulation,removal of free radicals and the promotion of energy metabolism and protein synthesis and,more importantly, can be a variety of traditional Chinese medicine compound preparation of the above three ways to achieve the cochlea improve the content of NT-3,which protect hair cells and auditory neurons and the role of auditory nerve fibers in order to reduce hearing damage.
