Objectives: This study was to investigate the expression of E2F-1,CyclinD1 and Rb in bladder transitional cell carcinoma (BTCC),and to analy respectively the relationship with clinicopathological factors(eg: tumor multiplicity, clinical stage, lymph node metastasis, pathological grade,et al.) and correlation between E2F-1, CyclinD1 and Rb. Explore the preliminary clinical significance of E2F-1, CyclinD1 and Rb in blader transitional cell carcinoma.Methods: Expression of E2F-1, CyclinD1 and Rb were evaluated by immunohistochemistry technique with 68 case of bladder transitional cell carcinoma and 18 normal bladder cases. The levels of the expression were analyzed combined with clinicopathological feature of patients including age, sex, tumor size, tumor multiplicity, loci of the tumor, lymph node metastasis, clinical stage, pathological grade, et al. Results: Buffy color staining found in nucleus was recognized as positive. E2F-1, CyclinD1 and Rb located in nucleus.1.The positive rate of E2F-1 and CyclinD1 in the 68 cases of bladder transitional cell carcinoma was 69.12%, 51.47%, respectively, which in the 18 cases of normal bladder tissue was 38.89%, 11.11% respectively. E2F-1 and CyclinD1 were highly expressed in bladder transitional cell carcinoma. A statistically significant differences of E2F-1 and CyclinD1 staining were observed between normal bladder tissues and bladder transitional cell carcinoma(p<0.01). The expression of Rb in the 68 cases of bladder transitional cell carcinoma was 48.53%, which in the 18 cases of normal bladder tissue was 100%. A statistically significant differences of Rb staining was observed between normal bladder tissues and bladder transitional cell carcinoma(p<0.01). 2. E2F-1 was highly expressed in bladder transitional cell cancer. But the rate of positive expression of E2F-1 in the bladder transitional cell carcinoma was not correlated with tumor grade(p>0.05), clinical stage (p>0.05) and tumor recurrence(p>0.05). CyclinD1 was highly expressed in bladder transitional cell cancer. And the rate of positive expression of CyclinD1 in the bladder transitional cell carcinoma was negatively correlated with tumor grade and clinical stage (p<0.01). Rb was lowly expressed in bladder transitional cell cancer. And the rate of positive expression of CyclinD1 in the bladder transitional cell carcinoma was negatively correlated with tumor grade and clinical stage (p<0.01).3.The rates of positive expression of CyclinD1 and Rb in the bladder transitional cell carcinoma were negatively correlated with tumor grade and clinical stage(p<0.01).But the rate of positive expression of E2F-1 was not significantly changed in the bladder transitional cell carcinoma (p>0.05).Conclusion:1. E2F-1,CyclinD1 and Rb might be involved in development of bladder transitional cell cancer.2. E2F-1 and CyclinD1 might played a important role in the earlier process of bladder transitional cell cancer. Rb might be closely related to invasive behavior of bladder transitional cell carcinoma.And it was a bad single for evaluation of prognosis.5. Both overexpression of E2F-1 and CyclinD1 and descended expression of Rb might be useful for early diagnosis.
