Objective:This study is a subsequent subject of"the standardize study on processing and fabrication of Rhizoma Typhonii", which is a sub-topic of the important tackle key problems projects of Henan Province"the standardize study on processing of traditional chinese medicinal(TCM ) slices". Rhizoma Typhonii origin in dry stem tuber of Tyhonium giganteum Engl.of Rhizoma Arisaematis which is from Henan Yuzhou.To compare the differences of ultraviolet spectrum and HPLC chromatogram of Rhizoma Typhonii and processed Rhizoma Typhonii, To isolate and identify the newly added chemical constituent from processed Rhizoma Typhonii,To establish qualitative and quantitative methods of the newly added chemical constituent from processed Rhizoma, To study the effect of different process methods on the content of the newly added chemical constituent from processed Rhizoma, To study antitumor effect of Rhizoma Typhonii, To study the effect of different processed products of Rhizoma Typhonii on tyrosinase activity, so as to provide scientific basis for the study on optimization of technology of processing,and offer application basis for development of new drug using anticancer activities and effectiveness and safety in Chinese clinical medicine.Methods: Comparison the differences of peak position and peak on ultraviolet spectrum group draft of different solvent site between Rhizoma Typhonii and processed Rhizoma Typhonii by Ultraviolet Spectral Line Group;Comparison the difference of chemical constituents between Rhizoma Typhonii and processed Rhizoma Typhonii by HPLC,and detection conditions of the newly added chemical constituent was determined by HPLC;using different extraction solvents and the newly added chemical constituent was determined by HPLC, Qualitative and separation conditions of the newly added chemical constituent was determined by TLC; The constituent were isolated and purified by column chromatography and preparative HPLC method, The structures were elucidated based on chemical and spectral analysis(UV,IR,1HNMR,13CNMR,MS);Glycoside from Rhizoma Typhonii was determined by molish reaction, Carbohydrate and the differences of the content of the newly added chemical constituent from Rhizoma Typhonii were analysis by HPLC-ELSD, Study the formation path of the newly added chemical constituent by simulated processing Rhizoma Typhonii; Determination of content of the newly added chemical constituent from different time and amount of alumen processing Rhizoma Typhonii by HPLC, Study the effect of different process methods on the content of the newly added chemical constituent from processing Rhizoma Typhonii by the Chinese pharmacopeia method, Study the effect of different process methods (pharmaceutical excipient amount, time, pressure, temperature) on the content of the newly added chemical constituent from processing Rhizoma Typhonii by pressure technology; Establish quantitative methods of the similar with the newly added chemical constituent by HPLC, and study the effect of different process methods on the content of the chemical constituent;Experimental study on anti-tumor activity of Rhizoma Typhonii in vivo, Observe the effect of water decoction and ethanol extract from Rhizoma Typhonii and processed Rhizoma Typhonii on the sarcoma grafts and immune organs of S180 mice; The relative inhibitory activity against tyrosinase of different processed products of Rhizoma Typhonii was detected by method of enzymatic activity determination.Results: The ultraviolet spectral line group of the raw product and the processed products showed distinctively different, The content of different solvent site from processed Rhizoma was reduced significantly. Compared with ethanol extraction, chloroform extraction and water extraction was reduced significantly, The two processed products are similar in the ultraviolet spectral line group; a newly added chemical constituent was found in processed products and its content was relatively higher than anyother; Tow compounds were isolated, The newly added chemical constituent is 5-hydroxym-ethyl-2-furfural, Its structure similar compound was bis(5-formyfuryl) ether;Rhizoma Typhonii mainly contains fructose, glucose, sucrose, etc. Sucrose can be transformed into 5-HMF at the processing Rhizoma Typhonii conditions; The content of 5-HMF from processing Rhizoma Typhonii by the Chinese pharmacopeia method increased with the heating time and amount of alumen in certain extent. The content of 5-HMF from processing Rhizoma Typhonii by pressure technology decreased with the temperature increasing and increased at first and then decreased with amount of alumen in certain extent, The content of 5-HMF was the highest at 9% alumen. The content of 5-HMF from Non-use of ginger products more than The use of ginger products; The content of bis(5-formyfuryl) ether showed: Rhizoma Typhonii
