Objective: Through literature review,market survey and origin survey of Rhizoma typhonii,the traditional classification method of traditional Chinese medicinal materials as a reference,combined with modern science and technology,the research on the specification grade and quality characteristics of Rhizoma typhonii medicinal herbs and decoction pieces was conducted.Standards provide scientific evidence.Method:Firstly,the characteristics of the 43 batches of samples collected were investigated,and the samples were classified according to the traits criteria,and 97 samples were obtained;secondly,based on the 2015 Chinese Pharmacopoeia,the moisture content,total ash content,alcohol-soluble extracts The ICP-MS method was used to detect Pb,As,Hg,Cu,Cd heavy metals and harmful elements in the sample.HPLC was used to analyze fructose,glucose,sucrose,maltose,maltotriose,cinnamic acid,urine,Pyrimidine,hypoxanthine,uridine,inosine,guanosine,thymidine,adenosine were measured.UV-visible spectrophotometer was used to determine the content of polysaccharide.Finally,correlation analysis and analysis of variance were performed on the experimental results.Results: The moisture content and total ash content of the sample were in compliance with the requirements of the 2015 edition of the Chinese Pharmacopoeia;the content of leachable white Rhizoma typhonii extracts was lower than that of the pharmacopoeia,and the leachables of other samples were qualified;the detection results of heavy metals and harmful elements showed that 29 batches of samples exceeded the standard,6 Batch Pb exceeded.The correlative analysis results showed that cinnamic acid was significantly negatively correlated with grades,thymidine was significantly positively correlated with grades,and fructose,uridine,guanosine,and adenosine were significantly positively correlated with grades.The results of variance analysis showed that there was a significant difference in the first and third grades of fructose,cinnamic acid,uridine,and thymidine in the herbs of Rhizoma typhonii,and there were significant differences in adenosine components between the second and the first and the third.In the guanosine composition,there was a significant difference between the second and third grades;in cinnamic acid,guanosine,and thymidine,there was a significant difference between the mildewed Rhizoma typhonii and normal white Rhizoma typhonii herbs;in fructose,glucose,sucrose,maltose,Maltotriose,cinnamic acid,polysaccharides,uracil,hypoxanthine,uridine,guanosine,thymidine,adenosine,and white Rhizoma typhonii were significantly different.Conclusion: The content determination method established in this study is stable,accurate,and reliable,and can be used as an evaluation index for the quality of white Rhizoma typhonii.Through the correlation study and variance analysis of grades with fructose,glucose,sucrose and other chemical components,it provides a theoretical basis for the classification of commercial specifications of the white monkshood.
